摘　　要：目的:运用高效液相色谱法测定道地产地秦艽药材中龙胆苦苷的含量。方法:色谱柱为Zorbax Extend-C18(150 mm×4.6 mm,5μm);流动相为甲醇-水(1∶4),流速为1 mL·min-1,检测波长为273 nm,柱温为40℃。结果:龙胆苦苷的线性范围为0.01～3.52μg,r=0.9999;仪器精密度的RSD为1.0%;方法重复性的RSD为1.5%;平均加样回收率为101.6%,RSD为3.0%(n=5)。结论:本法快速简便,重复性好,测定准确灵敏。秦艽(四基源药材)的种间龙胆苦苷含量有较大差异。Objective： To develop an RP - HPLC method for the determination of gentiopcroside in Radix Gentianae Macrophyllae （Qinjiao） in local area. Methods：A Zorbax Extend - C18 column （ 150 mm ×4.6 mm,5μm） was a- dopted the mobile phase of methanol - water （ 1 ： 4） at 40 ℃ with flow rate of 1.0 mL · min^-1, detecting wavelength being 273 nm. Results：The calibration curve was linear in the range of 0.01 -3.52 μg of gentiopcroside with cor- relation coefficient of 0. 9999. The instrument precision RSD was 1.0% and the method precision RSD was 1.5%. The average recovery was 101.6% （RSD = 3.0% ,n=5）. Conclusion：This method is simple and reproducible;the analytical results are reliable and accurate. The gentiopcroside contents are different among different species and sources of Radix Gentianae Macrophyllae.
