摘 要:目的:观察小剂量重复照射下不同时相大鼠血清内皮素(ET-1)和肿瘤坏死因子-α(TNF-α)的改变并探讨益气活血中药对放射性肺损伤的防治作用。方法:将96只W istar雌性大鼠随机分为照射加中药组(A组)30只、照射加氨溴索组(B组)30只、单纯照射组(C组)30只、正常对照组(D组)6只。用直线加速器对前三组大鼠右肺进行分次照射(5 Gy/次,1次/周,累积剂量最高为30 Gy),于照射开始后第4、6、8、12、26周5个时间点分别处死A、B、C组各6只大鼠,取血清标本进行ELISA检测,观察各组大鼠不同时相血清ET-1和TNF-α的动态变化。结果:3组血清ET-1水平于照射后4周进行性升高,8周达高峰,12周开始下降;3组血清TNF-α水平4周进行性升高,12周达高峰,26周开始下降。A组各时相大鼠血清ET-1、TNF-α表达水平均低于C组(P<0.05),且各时相值与B组近似,差异无统计学意义(P>0.05)。结论:血清ET-1、TNF-α水平的变化能反映早期放射性肺损伤的严重程度,益气活血中药能够降低血清ET-1、TNF-α的水平,可能是其防治放射性肺损伤的作用机制。Objective:To explore the changes of ET-1 ,TNF-α in plasma and the curative effect of Yiqi Huoxue Recipe in rats with radiation-induced lung injury in different radiation times. Methods:96 W istar rats were randomly divided into irradiation group:group A( n = 30 ) irradiation with Yiqi Huoxue Recipe group ; group B ( n = 30) irradiation with ambroxol group ; group C ( n = 30) irradiation group and group D (n = 6) normal control group. The first three groups were irradiated at right hemithorax with a dose of 30 Gy/6 Fraction/5 weeks. Animals were followed for 26 weeks after the first irradiation. The levels of serum ET-1 and TNF-α of 5 rats in each group were assayed by ELISA at the end of the 4th,6th,8th,12th and 26th week after the first irradiation. Results:The levels of serum ET-1 and TNF-α of the 3 groups increased progressively in the 4th week after irradiation, reached the peak in the 8th week and began to drop in the 12th week, but TNF-α level reached the peak in the 12th week and began to drop in the 26th week. The expression level of ET-1 and TNF-α were significantly lower than that of group C(P 〈0. 05) ,and in ambroxol-group,the levels of serum ET-1 and TNF-α were same to that of the Yiqi Huoxue Recipe group with no statistical significance( P 〉 0.05 ). Conclusion : ET-1 and TNF-α can reflect the degree of radiation-induced lung injury. Yiqi Huoxue Recipe can decrease the levels of ET-1 and TNF-α in plasma significantly. This might be one of the mechanisms in the treatment and prevention of radiation-induced lung injury.
