沙培林对U251胶质瘤细胞凋亡及相关基因表达的研究

摘　　要：目的研究沙培林对脑胶质瘤细胞凋亡及相关基因表达的影响。方法20只BALB/C裸小鼠皮下接种U251脑胶质瘤成功后随机分成4组,分别瘤周注射生理盐水0.2ml,沙培林0.2KE、0.4KE、0.8KE,共8次,解瘤实验。用免疫组化法检测Bcl-2、p53(突变型)、Bax、Fas及Bcl-xs基因的表达。结果由对照组至治疗组,且随用药剂量增加,Bcl-2及p53(突变型)表达的阳性率逐渐下降,而Bax、Fas及Bcl-xs表达的阳性率则逐渐增加。结论沙培林局部应用治疗脑胶质瘤,通过对多种基因的调控,促进瘤细胞的凋亡、抑制肿瘤的生长。Objective To investigate the effect of Sapylin to apoptosis and related gene expression of cerebral glioma. Methods 20 post vaccinial BALB/C nude mouse with U251 glioma were randomly divided into 4 groups. 0.2ml normal saline solution and 0.2 KE Sapylin and 0.4 KE Sapylin and 0.8 KE Sapylin were injected to periph- ery of tumor to respectively. After 8 times, the tumors were dissected. The gene expression of bcl - 2, p53, bax, fas and bcl - Xs was determined by immunohistochemical procedures. Results The results showed that the positive rates of bcl - 2 and mutational p53 were gradully decreased from control group to treated groups, and it was related to drug dose. But the positive rates of fas and bax and bcl - Xs were gradually increased. Conclusion Sapylin may induce glioma apoptosis and inhibit tumor growth through changing many genes.
