摘 要:目的观察电针对脑缺血大鼠额叶皮质缺血半影区Bax蛋白质表达的影响,为揭示电针治疗脑缺血的机制提供实验依据。方法雄性SD大鼠线栓大脑中动脉建立脑缺血动物模型,随机分为非电针组和电针组。用免疫组织化学的方法显示大鼠额叶皮质缺血半影区内Bax阳性细胞,观察形态和分布,并计算阳性细胞数目;用Western-blot方法检测非电针组和电针组大鼠额叶皮质缺血半影区Bax蛋白质表达量。将非电针组和电针组大鼠的同类指标与假手术组和正常对照组的同类指标比较,寻找组间差异。结果(1)免疫组织化学方法在对照组大鼠额叶皮质未显示出Bax免疫阳性细胞,在假手术组大鼠额叶皮质内也未见Bax免疫阳性细胞;非电针组大鼠额叶皮质缺血半影区于缺血6h即见圆形、椭圆形和梭形的Bax免疫阳性细胞,免疫阳性产物分布充满细胞质。电针组大鼠额叶皮质缺血半影区内Bax免疫阳性细胞在形态和分布上与非电针组未见明显组间差异。电针组的阳性细胞的数量比非电针组的明显减少,差异有统计学意义(P<0.01)。(2)Western-blot方法在对照组和假手术组大鼠额叶皮质内均未检出Bax蛋白质表达。电针组大鼠额叶皮质缺血半影区Bax蛋白质表达比非电针组的明显减少,差异有统计学意义(P<0.05)。结论2Hz频率电针刺激曲池和足三里穴位,可以下调脑梗死大鼠额叶皮质缺血半影区Bax蛋白质表达,参与脑缺血性神经元凋亡的病理过程。Objective To provide experimental evidence for studying mechanism of electroacupunture treating cerebral infarction.Methods Rat model of cerebral ischemia were established with the method of thread middle cerebral artery occlusion.56 the model were randomly divided into electroacupuncture group and non-electronacupuncture group.At the same time 7 control group rats and 7 sham group rats,which were only exposed middle cerebral artery without occlusion it,were matched.Immunochemistry method was used to show Bax positive cells in the cerebral cortex and the morphology and number of the positive cells were investigated and the level of Bax expression were assessed with Western-blot method.Results Bax positive cells were not found at frontal cerebral cortex in the control and sham group with immunohistochemistry.In nonelectroacupunture group Bax positive cells shwed after 6 h of ischemia at penambra area.Bax expression was peak at 1 day and at 3 days was less than 1 day,almost disappeared at 7 days.The morphology of positive cells in nonelectronacupuncture group was mainly round and ellipse or fusiform in shape,the distribution of the positive cells was mainly at the middle region of cortex.In electronacupuncture group,the morphology and distribution of positive cells were not significantly difference between two groups but the number of the positive cells and level of the Bax protein expression was decreased in electronacupunture group than non-electronacupuncture group.Conclusions Electonacupuncture on Neiguan and Zusanli acupoint can down regulate Bax protein expression at penambra area of cerebral cortex in middle cerebral artery occluded rats.
