摘 要:目的 建立虎杖中大黄素、大黄素甲醚含量的微乳液相色谱测定法。方法以微乳为流动相,色谱柱采用Hypersil BDS C185μm(4.6mm×150mm),检测波长为287nm,流速1ml/min,柱温30℃。外标峰面积法同时测定虎杖中大黄素和大黄素甲醚的含量。结果大黄素和大黄素甲醚的线性范围分别为5.3-53.0μg/ml(r=0.9999)和2.4~24.0μg/ml(r=1.0000),平均回收率和相对标准偏差(RSD)值(n=6)分别为97.7%,1.25%;97.6%,1.82%。结论本法简便快速精确,可用于虎杖中大黄素、大黄素甲醚的含量测定。Objective To establish the method for determination of emodin and physcion in Rhizoma et Radix Polygoni Cuspidati by microemulsion liquid chromatography (MELC). Methods The analytical column was Hypersil BDS C18 5μm (4.6 mm×150 mm), with column temperature of 30℃. The mobile phases was microemulsion which consisted of 4% w/v sodium dodecyl sulfate, 9% v/v butanol, 1.4% v/v octane and 0.5% triethylamine in water, flow rate was 1.0 ml/min, and detection wavelength was 287 nm. Results The calibration curve were linear with r =0.9999 in the rang of 5.3-53.0 μg/ml for emodin; r =1.0000 in the rang of 2.4-24.0 μg/ml for physcion. The average recovery and RSD (n =6) was 97.7%, 1.25%; 97.6%,1.82%, respectively. Conclusion The method is rapid, accurate, and reproducible, and can be used for the quality control of Rhizoma et Radix Polygoni Cuspidati.
