摘 要:以细胞转化为实验模型,应用细胞生物学和分子生物学的实验手段,研究了辐射所致人胚肺细胞体外转化以及辐射对细胞DNA单链断裂损伤效应的规律。接受0.25~5Gy60Coγ射线作用后第20代,各剂量组的人胚肺细胞半固体琼脂培养集落形成能力均明显增加,说明人胚肺细胞在射线作用下发生了转化,而且细胞形态转化的出现具有时相性及作用剂量依赖性。采用缺口翻译技术检测细胞DNA单链断裂损伤的结果表明,经不同剂量作用后,反映细胞DNA单链断裂损伤程度的3HdTTP掺入量,在各剂量作用组均明显增加,表明射线造成了细胞DNA损伤。DNA单链断裂损伤效应与剂量之间的关系亦表现为正相关。统计学分析表明,辐射所致细胞DNA单链断裂损伤与辐射诱发的细胞转化之间存在着相关关系,辐射诱发的细胞转化随着细胞DNA单链断裂损伤效应的加大而增强。这一结果提示,辐射致癌效应与DNA损伤效应密切相关。γ irradiation effects on transformation and DNA strand break of human embryo lung cell were studied by means of cell and molecular biology. The cell transformation appeared at 20 th passage post 0.25~5.0Gy γ irradiation. The transformation was relative to doses and post time of irradiation. DNA strand break was detected by NTA (nick translation assay) in this report. The result indicated DNA strand break was arisen from γ ray with different doses. DNA strand break also was relative to radiation doses as cell transformation. Correlation between DNA strand break and cell transformation was analysed. The analysis indicated that effect of cell transformation enhanced with increase of DNA strand break. It was suggested that carcinogensis by radiation is closely relative to DNA damage.
