摘 要:目的:探讨脂联素(ADPN)对体外高糖培养的人肾小管上皮细胞转化生长因子-β1(TGF-β1)表达的影响,以揭示ADPN对高糖环境中的肾小管上皮细胞的可能保护作用机制。方法:人肾小管上皮细胞分别培养于不同的培养基中,在高糖环境中加入脂联素,分别培育48h后采用酶联免疫吸附法(ELISA)测定细胞上清液中TGF—β1的含量。结果:TGF—β1在高糖组表达较高,加入中等浓度(ADPN浓度为10.0,15.0,20.0ug/mL)TGF—β1的表达减少,和高糖组相比,两者的差异有统计学意义。结论:中等浓度ADPN可以抑制TGF—β1在人肾小管上皮细胞的表达。Objective:To investigate the effect of adiponectin (ADPN)on the expression of transforming growth factor-β1 (TGF-β1)in human renal tubular epithelial cell cultured with high concentration of D-glucose. Methods:Human renal tubular epithelial cells were cultured in media containing high concentration of D-glucose or adiponectin. The expression of TGF-β1 was measured by enzyme-linked immunosorbent assay(ELISA)at the 48 h. Results :The expression of TGF-β1 was promoted in high glucose group. Compared with high glucose group ,the expression of TGF-β1 was suppressed in moderate concentration adiponectin(10.0,15.0,20.0 ug/mL). Conclusion:Moderate concentration adiponectincan decrease the expression of TGF-β1.
