摘  要:基于Chlorella pyrenoidosa的生长规律和在缺氮条件下培养可提高其细胞内的油脂积累的特性,进行了两步培养法提高小球藻油脂产量的研究.结果显示:小球藻高密度生长的最适葡萄糖和硝酸钾质量浓度分别为20 g/L和1.50 g/L;单步培养法中氮源KNO3初始质量浓度为1.50 g/L时的小球藻生物量为(7.55±0.23)g/L,两步培养法中的小球藻生物量提高至(9.23±0.11)g/L;与单步培养法相比,两步培养法中小球藻细胞的油脂含量和总脂肪酸含量分别提高了5.8%和5.2%,藻的热值由(23.58±0.02)kJ/g提高到(25.52±0.03)kJ/g.这说明,两步培养法不仅可保证藻细胞的高密度生长,而且能有效增加藻细胞的油脂含量.Based on the growth rules of Chlorella pyrenoidosa ( C. pyrenoidosa) and the characteristics of intracellular lipid accumulation in nitrogen-deficient condition, the improvement of lipid content of C. pyrenoidosa using a two-step cultivation method was investigated. The results show that ( 1 ) the optimal mass concentrations of glucose and KNO3 for the high-density cultivation of C. pyrenoidosa are respectively 20 and 1. 50 g/L; (2) with 1.50 g/L KNO3 as the nitrogen source, the biomass of C. pyrenoidosa increases from (7.55 ± 0. 23 ) g/L in the single-step cultivation to (9. 23 ±0. 11 ) g/L in the two-step cultivation; (3) the contents of intracellular lipid and total fatty acids in the two-step cultivation .respectively increase by 5.8% and 5.2% ; and (4) the calorific values improves from (23.58 ±0. 02) kJ/g to (25.52±0.03) kJ/g. It is thus concluded that the two-step cultivation not only results in a high-densitv cultivation of C. pyrenoidosa but also effectively increases the cellular linid content.
