左归丸与右归丸对EAE大鼠淋巴细胞亚群和NK细胞的影响

摘　　要：目的:通过观察左归丸与右归丸对大鼠实验性自身免疫性脑脊髓炎模型(EAE)的影响,了解两者作用机制的异同。方法:应用髓鞘碱性蛋白与完全福氏佐剂按体积比1 1制成抗原并于Lewis大鼠双后足垫下注射,建立EAE模型。通过流式细胞仪技术观察淋巴细胞亚群及NK细胞水平。结果:造模后第15天,左归丸组CD4+细胞显著升高(P<0.05);模型组CD4+/CD8+比值高于正常组(P=0.052)。造模后第27天,模型组CD4+/CD8+比值明显升高(P<0.05);左归丸组CD4+/CD8+比值明显降低(P<0.05);左归丸组和右归丸组NK细胞明显升高(P<0.05)。结论:EAE大鼠CD4+/CD8+比值升高,这在EAE发病中发挥重要作用。滋阴补肾法可上调急性期CD4+细胞水平,下调缓解期CD4+/CD8+比值来治疗EAE。左归丸与右归丸可下调急性期NK细胞水平,上调缓解期NK细胞水平治疗EAE。Objective： To observe the effect of ZuoGuiWan and YouGuiWan on induced experimental autoimmune encephalomyelitis （EAE） in Lewis rats, compare the therapeutic effect between ZuoGuiWan and YouGuiWan, and explore the underlying immunoregulatory mechanism between warming-yang and nourishing-yin therapy. Methods ： 120 male Lewis rats were bred at Capital Medical University under specific pathogen-free conditions. 24 rats were selected as normal group before immunization. The other rats were randomly devided into 4 groups after immunization, including EAE group, prednisone group, ZuoGuiWan group and YouGuiWan group. Rats of normal group were injected with normal sodium in the hind feet pad on both sides, the other rats were injected with the antigen consisted of myelin basic protein（MBP） and complete Freund＇s adjuvant（CFA） to induce EAE. Rats of normal group and EAE group were administered with normal sodium after immunization, rats of prednisone group with prednisone（5mg/kg）, rats of ZuoGuiWan group with ZuoGuiWan（2g/kg）, rats of YouGuiWan group with YouGuiWan（3g/kg）. On the day 15 and 27 after immunization, the peripheral blood was collected, lymphocyte subgroups and NK cells were detected by flow cytometer. Results： On day 15 after immunization： In peripheral blood, the number of CD4^＋lymphocyte in ZuoGuiWan group was significantly higher（P〈0.05） than that in normal group. The ratio of CD4^＋/CD8^＋ in EAE group was significantly higher（P〈0.05） than that in normal group. On day 27 after immunization： In peripheral blood, the ratio of CD4^＋/CD8^＋ in EAE group was significantly higher（P〈0.05） than that in normal group. The raito of CD4^＋/CD8^＋ in ZuoGuiWan group was significantly lower（P〈0.05） than that in EAE group. The number of NK cell in ZuoGui Pill group was significantly higher than normal group＇s, EAE group＇s and Prednisone group＇s（P〈0.01 with normal group and EAE group,P〈0.05 with Prednisone group）.
