摘  要:目的观察2型糖尿病大鼠心肌病变中心肌脂联素(adiponectin,APN)以及心肌脂联素受体1(adiponectin receptor1,AdipoR1)的表达情况,探讨心肌APN、AdipoR1的表达与糖尿病心肌病变的关系。方法23只雄性Wistar大鼠随机分为正常对照组(A组,n=10)和糖尿病组(B组,n=13)。用高糖高脂饲料加小剂量链脲佐菌素建立2型糖尿病大鼠模型。喂养3个月后应用免疫组织化学方法测定心肌AdipoR1的表达。用酶联免疫吸附法检测血浆和心肌APN水平。通过颈动脉插管测等容舒张期左室内压力下降最大速率(-dp/dtmax)和等容收缩期左室内压上升的最大速率(+dp/dtmax)。结果与A组比较,B组糖尿病大鼠3个月时出现心脏重量/体重水平升高(P<0.05),等容舒张期左室内压降低,血浆和心肌APN水平以及心肌AdipoR1表达水平降低(P<0.05)。结论2型糖尿病大鼠心肌APN及其AdipoR1表达水平的降低可能与糖尿病心脏肥大和心功能降低有关。Objective To observe the expression of cardiac adiponectin and its receptor-1 in type 2 diabetic rats, and to explore their relationship with diabetic cardiomyopathy. Methods Twenty-three male Wistar rats were randomized into control group ( n = 10) and diabetic group(n = 13). The rats were fed with a high-fat and high-sugar diet and injected with a low dose of STZ through tail vein to establish the type 2 diabetic rat models. After 3 months, cardiac expression of adiponeetin receptor 1 ( AdipoR1 ) was determined by immunohistochemistry. Plasma and myocardial adiponectin levels were assayed by enzyme immunoassay. The maximum descent-speed (-dp/dtmax) of pressure in left ventricle isovolumic relaxation period and the maximum ascent-speed( + dp/dtmax)of pressure in left ventricle isovolumic contraction period were measured through carotid artery cannula. Results After 3 months, the ratio of heart weight to body weight in diabetic rats significantly increased (P 〈 0.05 ). The heart function, plasma and myocardial adiponectin levels, and the protein expression of myocardial adipoR1 significantly decreased in diabetic rats compared with that in controls (P 〈 0.05 ). Conclusion The decreased myocardial expression of adiponeetin and its receptor 1 may be related to the cardiac enlargement and the decreased heart function in type 2 diabetic rats.
