摘  要:目的记录双电极绑定条件下多组高频刺激(HFSs)诱导的大鼠在体海马CA1区长持续长时程增强(L-LTP)现象。方法雄性Wistar大鼠乌拉坦麻醉后固定于脑立体定位仪上。埋置脑室导管并安装绑定的刺激/记录电极。引导基础性场兴奋性突触后电位(fEPSP)和施加多组HFSs诱导L-LTP。结果绑定后的刺激和记录电极能稳定地引起海马CA1区fEPSP。基础性fEPSP记录可保持长时间稳定。有效的多组HFSs可有效诱导至少维持3h以上的L-LTP,成功率高达65%。脑室注射β淀粉样蛋白(Aβ)对L-LTP的维持显示出明显的压抑作用。结论采用绑定的刺激/记录电极进行L-LTP记录简便易行。利用多组HFSs方案诱导L-LTP成功率较高。Objective To record the late-phase long-term potentiation (L-LTP) in the CA1 area of rats in vivo by using electrodes-binding technique and delivering multiple high frequency stimulations(HFSs). Methods Wistar rats were anesthetized with urethane and fixed with the stereotaxic device for acute surgery. Cannula was implanted into lateral cerebral ventricle,and bound stimulating/recording electrodes were inserted into hippocampal CA1 area. Basal field excitatory postsynaptic potential(fEPSP)and multiple HFSs-induced L-LTP were recorded. Results fEPSPs were steadily induced by using the electrodes-binding technique. Basal fEPSP recording was very stable, lasting for a long period enough to finish the whole experiment. The multiple HFSs successfully induced L-LTP, with a higher incidence up to 65% and a longer maintenance for at least three hours. Intracerebroventricular( i. c. v. ) injection of amyloid beta(AI3) suppressed the maintenance of HFSs-induced L-LTP significantly. Conclusion The electrodes-binding technique for recording hippocampal L-LTP in vivo is quite simple and convenient. The rates of fEPSP appearance and L-LTP induction are kept high by delivering multiple HFSs.
