摘 要:目的探讨脂联素(ADPN)对体外高糖培养的人肾小管上皮细胞增殖及细胞间黏附分子1(ICAM-1)表达的影响,揭示脂联素对高糖环境中的肾小管上皮细胞可能的保护作用机制。方法①将人肾小管上皮细胞用5.5mmol/L葡萄糖(正常对照组)、30.0mmol/L葡萄糖(高糖组)、5.5mmol/L葡萄糖+24.5mmol/L甘露醇(甘露醇组)、30.0mmol/L葡萄糖+2.5μg/mlADPN(脂联素组)分别培养24h,48h,96h后,运用MTT法测定细胞增殖。②将人肾小管上皮细胞用5.5mmol/L葡萄糖(正常对照组)、30.0mmol/L葡萄糖(高糖组)、30.0mmol/L葡萄糖+不同浓度ADPN(ADPN终浓度分别为2.5,5.0,7.5,10.0,15.0和20.0μg/ml,脂联素组)分别培养48h后,运用酶联免疫吸附法(ELISA)测定细胞上清液中ICAM-1的含量。结果①高糖组细胞增殖被抑制,加入低浓度脂联素(2.5μg/ml)后促进了细胞增殖;②ICAM-1在高糖组表达较高,加入中等以上浓度脂联素(ADPN浓度为10.0,15.0,20.0μg/ml)ICAM-1的表达减少。结论结果提示脂联素可能通过减少ICAM-1的表达而起到保护肾脏的作用。Objective To explore the protective effects of adiponectin(ADPN) on human renal tubular epithelial cells under the condition of high concentration of D-glucose. Methods (1)The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose ( control group), 30.0 mmol/L glucose ( high glucose group ), 5.5 mmol/L glucose + 24.5 mmoL/L mannitol ( mannitol group), 30.0 mmol/L glucose + 2.5μg/ml adiponectin ( ADPN group), respectively. After culture for 24 b, 48 h, 96 h, the proliferation activity of human renal tubular epithelial cell was observed by MTT.(2)The human renal tubular epithelial cells were cultured with 5.5 mmol/L glucose( control group) ,30.0 mmol/L glucose( high glucose group) ,30.0 mmol/L glucose + different concentrations of adiponectin(2.5, 5.0,7.5,10.0,15.0,20.0 μg/ml, ADPN groups), respectively. The expression of intercellular adhesion molecule-1 ( ICAM-1 ) was measured by enzyme-linked immunosorbent assay(ELISA)after culture for 48 h. Results The proliferation of human renal tubular epithelial cells decreased in high glucose group, but increased in 2.5 μg/ml adiponectin group. The expression of ICAM-1 increased in high glucose group,but decreased in 10.0,15.0,20.0 μg/ml adiponectin groups. Conclusion The results indicate that adiponectin maybe protect the kidney in patients with diabetes mellitus by decreasing the expression of ICAM-1.
