摘 要:目的观察骨髓来源干细胞(BMDSCs)能否在体分化为其他实体组织细胞。方法野生型C57BL/6J雌性小鼠作为受体接受10Gy的射线照射后,经尾静脉植人同等背景的转增强型绿色荧光蛋白(EGFP)基因的C57BL/6J雄性小鼠(绿鼠)骨髓细胞1×10^7个/只。移植受体稳定1年后检测各组织中EGFP的表达。结果野生型小鼠各组织中EGFP的表达为0%,绿鼠各组织中EGFP的表达为100%。受体鼠各组织均有EGFP阳性细胞分布,表达率为100%,与野生型比较差异有统计学意义(P〈0.01),但是表达强度均低于绿鼠组。EGFP阳性细胞主要存在于皮肤组织角质形成细胞、毛囊上皮,以及肺间质、支气管上皮、肺泡上皮中。结论BMDSCs能在体内分化为其他实体细胞,并有组织特异性。Objective To study wheather bone marrow-derived stem cells (BMDSCs) can differentiate to somatic cells in vivo. Methods Wild-type C57BL/6J female mice underwent 10 Gy radiation exposure,then received 1 ×10^7 bone marrow cells each from male C57BL/6J mice ( green mice) which were transfected with enhanced green fluorescent protein (EGFP). One year later the recipients were sacri- riced and individual tissues were investigated for the expression of EGFP by fluorescence and immunohistochemistry. Results The EGFP expression was 0% in Wild-type mice, but 100% in green mice. EGFP- positive cells were found in tissues of bone marrow recipients, including the skin, liver, lung, kidney and heart, especially greater in follicle epithelial ceils and kerotinocytes of the skin, and epithelial ceils of bronchus and alveolus, as well as interstitial tissue. Conclusion BMDSCs could trandifferentiate into somatic cells and the process was characterized by stimuli of injury.
