摘  要:目的观察雌性大鼠尿道括约肌损伤修复过程中内源性胰岛素样生长因子表达。方法制作气囊扩张模拟产伤的SD大鼠尿道括约肌损伤模型,分别于损伤后2、4、6、8、14d处死大鼠取出尿道,逆转录-聚合酶链反应(RT—PCR)法测定IGF—Ⅰ和Ⅱ的mRNA表达,尿道括约肌无损伤大鼠为对照组。结果IGF—ⅠmRNA在损伤后4、6、8、14d表达,尤以第8天升高明显(P〈0.05);IGF-ⅡmRNA在伤后4、6、8d表达,以第6、8天升高明显(P〈0.05);未损伤组无明显IGF表达。结论内源性IGF—Ⅰ和Ⅱ参与尿道括约肌损伤修复过程,提示IGF对尿道括约肌损伤具有潜在治疗作用。Objective To investigate the expression of insulin-like growth faetor-Ⅰ and Ⅱ( IGF- Ⅰ and Ⅱ ) during healing after the urethral sphincter muscle injury in female Sprague-Dawley (SD) rats. Methods The model of urethral sphincter muscle injury in female rats ( n = 25) was made by intravaginal balloon inflation. The rats were killed at 2,4,6,8,14 days after trauma respectively, and the urethral specimens were harvested and processed for RT-PCR. A control group (n = 5) underwent the same procedure without intravaginal balloon inflation. Results The expression of IGF- I mRNA was increased at day 4,8, and 14 after urethral sphincter muscle injury, especially at day 8 ( P 〈 0. 05 ) ;The expression of IGF- Ⅱ mRNA was increased at day 4,6,8 after urethral sphincter muscle injury,especially at day 6 and 8 (P 〈 0. 05) ; No expression of IGF-Ⅰ and Ⅱ mRNA was detected in the control group. Conclusion IGF-Ⅰ and Ⅱ mRNA was expressed during healing after the urethral sphincter muscle injury and related to the healing process. IGF- Ⅰ and Ⅱ probably play a role in the treatment of stress urinary incontinence induced by urethral sphincter muscle dysfunction.
