摘  要:目的:观察血管紧张素-Ⅱ(Ang-Ⅱ)对血管内皮细胞(VEC)游离钙离子浓度([Ca2+]i)的影响及阿托伐他汀的保护作用。方法:将血管内皮细胞分为空白对照组(仅给予细胞培养液)、Ang-Ⅱ组(细胞培养液中加入Ang-Ⅱ,使其终浓度为10-7mol/L)、Ang-Ⅱ+小剂量阿托伐他汀组(在单纯Ang-Ⅱ组的基础上加入阿托伐他汀,使阿托伐他汀的终浓度为0.1μmol/L)、Ang-Ⅱ+大剂量阿托伐他汀组(在单纯Ang-Ⅱ组的基础上加入阿托伐他汀,使阿托伐他汀的终浓度为1μmol/L)。用激光共聚焦显微镜测量各组细胞的[Ca2+]i。结果:(1)Ang-Ⅱ组的[Ca2+]i显著高于空白对照组(P<0.01);(2)Ang-Ⅱ+阿托伐他汀组的[Ca2+]i显著低于Ang-Ⅱ组(P<0.01)。(3)Ang-Ⅱ+大剂量阿托伐他汀组的[Ca2+]i低于Ang-Ⅱ+小剂量阿托伐他汀组(P<0.05)。结论:Ang-Ⅱ可引起细胞内[Ca2+]i的显著增加,阿托伐他汀对Ang-Ⅱ所致的[Ca2+]i增加具有保护作用,且这一作用呈剂量依赖性。Objective To investigate the effect of Ang-Ⅱ on intracellular free calcium concentration([Ca^2+]i) in vascular endothelial cells (VEC) and the protective effect of atorvastatin. Methods VEC were divided into 4 groups: control group (only given cell culture medium); Ang-Ⅱ group (10^-7 mol/L Ang-Ⅱ ); Ang-Ⅱ + low dose atorvastatin group (0.1 μmol/L atorvastatin, 10^-7 mol/L Ang-Ⅱ ) ; Ang-Ⅱ + high dose atorvastatin group (1 μmol/L atorvastatin, 10^-7 mol/L Ang-Ⅱ ). [Ca^2+]i were measured by laser scanning confocal microscopy in the four groups. Results [Ca^2+]i in Ang-Ⅱ group was significantly higher than those in control group (P 〈 0.01 ), Ang-Ⅱ + low dose atorvastatin group (P 〈 0.01), and Ang-Ⅱ + high dose atorvastatin group (P 〈 0.01 ). [Ca^2+]i in Ang-Ⅱ + high dose atorvastatin group was significantly lower than that in Ang-Ⅱ + low dosage group(P 〈 0.05). Conclusion Ang-Ⅱ can increase [Ca^2+]i in VEC, which can be pretected by atorvastatin in a dose-dependent manner.
