山羊-绵羊异质克隆胚在G1/G2培养液中发育的可能性

摘  要:以山羊皮肤成纤维细胞为供体细胞,绵羊去核卵母细胞为受体细胞进行种间体细胞核移植,构建山羊-绵羊重构胚,并采用G1/G2培养液对其进行体外培养,比较G1/G2培养不同阶段添加饲养层对山羊-绵羊种间体细胞核移植重构胚发育的效果。结果表明,共构建获得山羊-绵羊重构胚472枚,重构胚能够在G1/G2连续培养基中发育至囊胚阶段;仅在G1中添加饲养层与对照组不添加饲养层相比,前者能获得较好的分裂率和桑囊率,差异不明显(卵裂率为76.88%vs 67.11%,桑椹胚率为52.84%vs 39.22%,囊胚发育率为7.32%vs 5.88%),而仅在G2阶段共培养与对照组相比培养桑囊率较低,差异不显著(卵裂率为69.38%vs67.11%,桑椹胚率为37.84%vs 39.22%,囊胚发育率为3.60%vs 5.88%),即,对于绵羊-山羊重构胚的培养,在前期(G1中)添加颗粒细胞培养重构胚效果优于仅在后期共培养(卵裂率为76.88%vs 69.38%,桑椹胚率为52.84%vs 37.84%,囊胚发育率为7.32%vs 3.60%)。Totally 472 heterogeneous embryos were reconstructed with goat skin fibroblasts as the donor cells and sheep enucleated oocytes as the recipient, and the effects of two culturing ways, which were respectively cultured in G1 added with granulosa cell, not in G2,and cultured not in Gladded with granulosa cell, but in G2. The result indicated that a total of 472 heterogeneous embryos were reconstructed, and could develop into blastocysts in G1/G2 in both ways, and in the former way of adding granulosa cell simply in the stage of G1, we got the better result compared with the control (cleavage rates were 76.88% vs 67. 11%, morulas rates were 52.84% vs 39.22%, and blastocysts rates were 7.32%vs 5.88%), while worse results were conducted in the latter means (cleavage rates were 69.38%vs 67.11%, morulas rates were37.84%vs 39.22 %, and blastocysts rates were 3.60% vs 5.88%). That was, it is better for goat-sheep heterogeneous embryos that the reconstructed embryos were cultured in G1 added with granulosa cell, not in G2, than that in G2 with granulose cell, not in G1 (cleavage rates were 76.88%vs 69.38%, morulas rates were52.84%vs 37.84%, and blastocysts rates were 7.32%vs 3.60%).
