冠心Ⅱ号不同配伍对丹参素血药浓度的影响

摘　　要：目的:考察冠心Ⅱ号方中君、臣、佐、使药不同配伍对丹参素血药浓度的影响。方法:采用高效液相色谱法测定丹参素血浆浓度,在煎煮条件一致的情况下,测定不同时间下冠心Ⅱ号方中君、臣、佐、使药不同配伍的煎剂灌胃大鼠后丹参素血浆浓度。结果:大鼠灌胃丹参单煎液、丹参川芎赤芍煎液后血浆中未测出丹参素;灌胃丹参川芎赤芍红花煎液后丹参素的血药浓度较低;灌胃丹参川芎煎液、冠心Ⅱ号全方煎液后丹参素的血药浓度较高,且血药浓度峰值时间从120min提前到90min。结论:君药川芎、臣药红花和佐使药降香可提高丹参素的血药浓度,臣药赤芍可降低丹参素的血药浓度。OBJECTIVE：To investigate the effects of the addition of different drugs （principal drug, ministerial drug and adjuvant and messenger drug） to Guanxin Ⅱ on plasma concentration of danshensu.METHODS： The plasma concentration of danshensu was determined by HPLC.Under the same decocting condition, the rat plasma concentration of danshensu was determined after intragastric administration of the decoctions of Guanxin Ⅱ in combination with different drugs（principal drug, ministerial drug and adjuvant and messenger drug） .RESULTS：The plasma concentration of danshensu in rats was failed to be detected after intragastric administration of decoction of single Salvia rniltiorrhiza or the decoction of Salvia miltiorrhiza, Ligusticum chuanxiong and Radix Paeoniae Rubra .The plasma concentration of danshensu in rats was low after administration of the decoction of Salvia miltiorrhiza, Ligusticum chuanxiong, Radix Paeoniae Rubra and Carthamus tinctorius. However, high plasma concentration of danshensu was noted in rats after administration of the decoction of Salvia miltiorrhiza and Ligusticurn chuanxiong or the whole formula decoction of Guanxin Ⅱ ,and the peak plasma concentration time was shortened to 90 min from 120 min. CONCLUSION ： The principal drug Ligusticurn chuanxiong, ministerial drug Carthamus tinctorius and adjuvant and messenger drug Dalbergia odorifera contributed to the increase of the concentration of danshensu, while which could be decreased by the ministerial drug Radix Paeoniae Rubra.
