止血固脱方对免疫低下小鼠T细胞亚群及IL-2水平影响的实验研究

摘　　要：目的:探讨止血固脱方对免疫低下小鼠T细胞亚群及IL-2水平的影响。方法:选取(20±2)g昆明种小鼠40只,随机分为空白对照组、环磷酰胺组、止血固脱方大剂量组(大剂量组)、止血固脱方小剂量组(小剂量组)4组,每组10只。除空白对照组外,其余各组小鼠每只按3mg/(kg·d)剂量腹腔注射环磷酰胺0.2mL,每天给药1次,连续7d,造成免疫低下小鼠模型。止血固脱方大、小剂量组分别按100g/(kg·d)、10g/(kg·d)剂量给药,2次/d,连续给药7d。最后一次给药24h后,采用断头采血法,每只采血0.4mL,将0.2mL加入抗凝试管中,采用流式细胞仪检测T细胞亚群,0.2mL放入离心机,以2000转/min离心20min,取血清速冻,采用放免γ测量仪检测IL-2含量。结果:止血固脱方能显著增加注射环磷酰胺引起的免疫低下小鼠的CD+4T细胞亚群的数量,增高CD+4/CD+8比值,提高血清中IL-2生成水平。结论:止血固脱方治疗过敏性紫癜(肾型)的免疫学机理可能与调节免疫系统功能紊乱有关。Objective： To observe the effects of Zhixue Gutuo Decoction on the contents of T lymphocyte subset and IL-2 of hypoimmunity model mice. Methods ：40 mice were randomly divided into control group,model group,Zhixue Gutuo Decoction small dosage group and large dosage group. Hypoimmunity model mice were made by intraperitoneally injecting cyclophosphamide once a day for successive 7 d. At the end of treatment, all mice were killed, then the percentages of CD4, CD8 in peripheral plasma of mice were detected by flow cytometry and IL-2 content in serum was detected by radioimmunoassay. Result： Compared with model group,the contents of CD4, IL-2 and the ratio of CD4 to CD8 in treatment group mice were significantly increased. Conclusion ： Therapeutic effect of Zhixue Gutuo Decoction on Henoch-Schonlein purpura nephritis may be associated with regulating the immune function.
