益气养阴活血化淤方对实验性病毒性扩张型心肌病小鼠细胞凋亡因子Fas/FasL蛋白表达的影响

摘　　要：目的通过观察小鼠心肌细胞凋亡因子Fas/FasL蛋白表达的水平,探讨益气养阴、活血化淤方的作用机制及Fas/FasL系统在细胞凋亡中的重要作用。方法210只Balb/c小鼠,随机分为4组。空白对照组30只;模型组、中药高剂量组、中药低剂量组各60只,采用多次接种柯萨奇B3m病毒的方式,建立重复感染的早期病毒性扩张型心肌病动物模型,中药高剂量组、中药低剂量组用益气养阴、活血化淤方治疗,于治疗4周后观测小鼠心肌病理形态学改变,免疫组化方法检测Fas/FasL蛋白表达。结果模型组心肌组织中Fas/FasL蛋白的阳性表达较空白组增多(P<0.05),中药高剂量组和中药低剂量组较模型组显著降低(P<0.05)。结论益气养阴,活血化淤方能够下调扩张型心肌病小鼠Fas/FasL蛋白表达,对心肌细胞起到保护作用。Objective To investigate the effect of the prescription of Yi Qi Yang Yin Huo Xue Hua Yu on the expression of myo- cardial Fas/Fasl protein in viral ditated cardiomyopath mice. Methods 210 Balb/c mice were randomly divided into 4 groups： blank group, model group, high dose traditional Chinese medicine group and low dose traditional Chinese medicine group. The mice of blank group were given relative dilute solution. The others were injected the CVB3m virus to the peritoneal cavity to set up the animal model of the viral dilated cardiomyopathy. Then the relative medicine was given for 30 days after 3 months. The immunohistochemical method was used to detect the expression of Fas/FasL protein in cardiac muscle. Results The expression of Fas/ FasL protein in cardiac muscle in model group was higher than that in blank group. The expression of Fas/FasL protein in cardiac muscle in high dose traditional Chinese medicine group and low dose traditional Chinese medicine group were significantly lower than that in model group. There was no significant difference between the high dose traditional Chinese medicine group and low dose traditional Chinese medicine group. Conclusion The prescription of Yi Qi Yang Yin Huo Xue Hua Yu can inhibit apoptosis of myocardial cells. The mechanism is that the prescription of Yi Qi Yang Yin Huo Xue Hua Yu can protect the cardiac muscle by reducing the expression of Fas/ FasL protein .
