摘  要:目的研究我国石斛属植物问的亲缘关系。方法从55个ISSR引物中筛选出14个多态性好的引物对石斛属(Dendrobium)植物基因组DNA进行简单重复问序列(ISSR)分析。利用POPGENE32软件计算ISSR扩增产物的Neig基因多样性指数(H)和Shannon信息指数(I),标记系统的有效等位基因数(effective number of alleles,NE)。应用NTSYS(3.0)软件计算Neig遗传距离和遗传相似系数。按照非加权平均距离法(UPGMA)构建聚类关系图。结果14条引物共扩增出123条带,多态性带112条,约占总数的91.06%,平均每个引物扩增的DNA带数为8.79条。Neig基因多样性指数(H)为0.3519,Shannon信息指数(I)为0.5282,有效等位基因数(NE)为1.5945,种间的相似系数介于0.0909~0.6957,平均为0.3779。结论我国石斛品种的遗传基础比较丰富,ISSR分子标记构建的亲缘关系树状图与经典分类系统基本一致.Objective To probe the relationship among Dendrobium species (Orchidaceae). Methods 14 primers with good pol- ymorphism screened from 55 ISSR primers were used to conduct ISSR analysis and dimorphic data of DNA fragments were used to construct the dendrogram by UPGMA. Results Totally 123 bands were amplified and 112 of them were polymorphic accounting for 91.06%. The average number of DNA bands amplified by each primer was 8. 79. Nei's gene diversity (H), Shannon's informa- tion index (I) and effective number of alleles(NE) were 0. 351 9, 0.528 2 and 1. 594 5, respectively. The genetic similarity a- mong all the tested clonal cuhivars ranged from 0. 090 9 to 0. 695 7, averaging 0. 377 9, which indicated that the genetic basis was relatively rich. Conclusion Clustering results revealed that the dendrogram constructed with ISSR molecular makers was iden- tical with the classic taxonomic system.
