解郁1号对抑郁大鼠血浆促肾上腺皮质激素释放因子和皮质醇含量的影响

摘　　要：目的探讨以调理脾胃法组成的中药复方解郁1号对抑郁大鼠血浆促肾上腺皮质激素释放因子、皮质醇含量的影响。方法SD大鼠40只,按随机表随机分为正常组、模型组、阿米替林组、解郁1号12,6 g/(kg.d)剂量组,每组各8只。除正常组外,各组接受21 d长期慢性刺激,在造模同时即开始给药。各组动物末次造模结束后,采用旷场实验进行行为学评分;采用放射免疫法测定促肾上腺皮质激素释放因子、皮质醇含量。结果实验大鼠38只进入结果分析。①各组大鼠血浆皮质醇含量的变化:模型组与正常组比较,血浆皮质醇水平明显升高(P<0.01)。中药高、低剂量组、西药组,与模型组相比,明显降低,具有显著差异性(P<0.01),但三组之间相比较,均没有统计学意义(P(0.05)。②各组大鼠血浆促肾上腺皮质激素释放因子含量的变化:模型组大鼠血浆的促肾上腺皮质激素释放因子含量高于正常组(P<0.01);与模型组比较,中、西药治疗组促肾上腺皮质激素释放因子含量多明显降低(P<0.01);中、西药治疗组之间促肾上腺皮质激素释放因子含量比较未见显著性变化(P(0.05)。结论解郁1号具有抗抑郁作用,其作用机制可能通过调节下丘脑-垂体-肾上腺轴(HPA),来调节血浆内促肾上腺皮质激素释放因子、皮质醇水平,达到抗抑郁目的。Objective To investigate the effect of JieYu No. 1 Decoction （JY）, which was a complex prescription with the function of adjusting spleen and stomach , on the contents of CORT and CRF in blood plasma of depression rats. Methods Isolated - living condition and chronic mild unpredictable stressure were applied suceessively to set up DEP rat model. Radiative immunotherapy was used to measure the contents of CORT and CRF in blood plasma. Results The results showed that the contents of CORT and CRF in blood plasma of depressed animals were much higher than those of normal animals （P 〈 0.01 ）. JY Decoction could descend the contents of CORT and CRF, and there was no significant difference between control group with normal and positive group（P 〉 0.05 ）. Conclusion JY Decoction can inhibit the occurrence of depression, and its mechanism may be related to descend the contents of CORT and CRF in blood plasma.
