摘 要:目的了解缺锌对睾丸发育及其功能的影响,研究缺锌对睾丸生精细胞凋亡的影响。方法选用Wistar雄性大鼠16只,均分为对照组和缺锌组。喂养4周后,采用原子吸收分光光度法,测定血清和睾丸锌的含量;采用原位缺口平移技术观察睾丸曲细精管中细胞凋亡数目的变化。结果在缺锌组大鼠血清锌(16.4±0.8μmol/L)较对照组血清锌(21.6±1.4μmol/L)明显降低的情况下,锌缺大鼠睾丸内锌含量(39.7±10.8μg/g)也较对照组(54.4±12.4μg/g)明显降低,睾丸曲细精管生精细胞凋亡的数目(21.4±3.8No./10个曲细精管)则较对照组的凋亡细胞数目(6.9±1.3No./10个曲细精管)明显增多。结论锌是睾丸发育所必需的营养素,缺锌可诱发睾丸生精细胞凋亡。Objective To study the changes of testis apoptosis in zinc deficient rats will promote the understanding of the molecular mechanism of zinc deficiency in the development and function of testis. Methods 16 Wistar rats were divided randomly into zinc control group (ZC) and zinc deficiency group (ZD). The serum and testis zinc contents were measured with atomic absorse method; the apoptosis of spermatogenic cells was studied with in situ nick translation (ISNT) technique. Results Under zinc deficient status, the zinc contents of the serum and testis were obviously decreased ( P <0 05). The apoptosis number of spermatogenic cells was significantly increased ( P <0.01). Conclusion The adequate amount of zinc is essential to the development of testis, whereas zinc deficiency can harmfully affect it. This effect is perhaps carried out in different ways, but the increasing apoptosis numbers of spermatogenic cells might be one of molecular miechanisms of the effect of zinc defficiency on testis development.
