摘　　要：目的研究叉状头转录因子Foxp3在变应性鼻炎小鼠模型外周血单个核细胞(peripheral blood monocyte,PBMC)中的表达并与正常小鼠比较。方法选取8周雌性BALB/c小鼠30只,分为实验组和对照组,每组15只。卵清蛋白(ovalbumin,OVA)致敏激发,建立BALB/c小鼠变应性鼻炎模型。21d后处理动物,取鼻腔固定脱钙后染色,观察其形态学特点;取脾组织研磨后红细胞裂解液分离获得PBMC,提取RNA行逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR),检测Foxp3mRNA表达情况。Foxp3相对表达量采用Foxp3与内参照GAPDH灰度比值表示。结果与对照组相比,实验组鼻腔黏膜部分脱落,黏膜下可见浆液腺增生和嗜酸性粒细胞浸润。Foxp3/GAPDH灰度比,实验组Foxp3mRNA表达明显低于对照组,差异有统计学意义(P<0.05)。结论Foxp3mRNA在变应性疾病中表达下调,低于正常对照,提示Foxp3作为CD4+CD25+Treg细胞的特异性活化标志,在变应性疾病的发病机制中可能起重要调节作用。Objective To investigate the level of transcription factor Foxp3 rhinitis level in the peripheral blood monocyte （PBMC） of mice with allergic rhinitis, and its relationship with pathogen of allergic AR. Methods 30 female BALB/c mice were divided into 2 groups：ovalbumin（ OVA）group and control. After 21 days the AR models were established. The pathological differences between the two groups were then surveyed. Reverse transeription-polymerase chain reaction （RT-PCR）was used to examine Foxp3 mRNA expression in the spleen PBMC from the two groups. Results In contrast to the control, OVA models had significant pathological changes in the nasal cavity, including epithelial damage and eosinophil infiltration in the submueosa. Foxp3 mRNA expression was lower in subjects with AR than that in the controls （ P 〈 0.05 ）. Conclusion The expression of Foxp3 mRNA in the OVA group is low which suggests that Foxp3 may play an important regulatory role in the pathogenesis of AR.
