摘  要:[目的]用携带BMP-2基因的腺病毒表达载体(Ad—BMP一2)转染兔骨髓基质干细胞(MSC),接种于胶原基纳米骨支架(nHAC)体外构建组织工程骨。[方法]将BMP-2基因修饰的MSC接种于nHAC支架,扫描电镜观察细胞在支架材料中的黏附、生长状况;WesternBlot分析BMP-2基因表达。[结果IBMP-2基因修饰的MSC在nHAC支架表面和孔隙内黏附、生长良好,仍可高水平表达BMP-2蛋白。[结论]BMP-2基因修饰的MSC在nHAC支架中黏附、生长良好,继续高水平表达并分泌BMP-2,组织工程骨构建成功。[Objective] To construct tissue-engineered bone via bone marrow stromal cells (MSC) which were transferred by adenovirus expression vector with bone morphogenetic protein-2(BMP-2) were planted into the scaffold of nano-hydroxyapatite collagen (nHAC). [Methods]The adhesion and growth of MSC modified by BMP-2 gene on the nHAC scaffold were observed by scanning electronic microscopy (SEM). Western blot analysis was used to determine the expression of BMP-2 protein. [Results] MSC modified by BMP-2 gene adhered to the surface and hole of the nHAC scaffold and grew well by SEM observation. The expression of BMP-2 was confirmed by Western blot analysis. [Conclusion] nHAC scaffold is in favor of the adhesion, growth and proliferation of trans gene cells. Trans-gene cells may still express BMP-2 highly. Tissue-engineered bone for regional gene therapy is constructed successfully.
