摘  要:[目的]初步研究肥胖候选基因过氧化物酶增殖激活物受体-γ2(PPAR-γ2)基因Plo12Ala的变异以及解偶联蛋白2(UCP2)基因Ala55Val的变异与成都地区人群单纯性肥胖的关系,并考察两者是否存在协同作用,从而为成都地区单纯性肥胖的研究与防治提供参考。[方法]将研究对象分为单纯性肥胖组和对照组。用聚合酶链反应—限制性片断长度多态性的方法(PCR-RFLP)检测两组研究对象的PPAR-γ2、UCP2基因的变异情况,分析两个基因的变异以及联合变异在两组间差异是否具有统计学意义。[结果]UCP2基因的变异在两组中的差异有统计学意义(P﹤0.05);而PPAR-γ2基因变异以及两者的联合变异在两组中的差异无统计学意义(P﹥0.05)。[结论]UCP2基因Ala55Val变异可能与成都地区单纯性肥胖相关,而PPAR-γ2的Plo12Ala基因的变异不是成都地区人群肥胖的危险因素,且两者不具有协同作用。[Objective] To tentatively study the peroxisome proliferator-activated receptor γ2 and uncoupling protein 2 gene polymorphism with simple obesity population in Chengdu, as well as the combined effects of the two variations, which would provide the information for the research and prevention of simple obesity in Chengdu. [Methods] Subjects were divided into 2 groups, including simple obesity group and the control. PCR-PFLP was applied to detect the PPAR-γ2 gene Plo12Ala and UCP2 gene Ala55Val variations. The variations of the PPAR-γ2, UCP2 and association of the two genes’ variations among the 2 groups were analyzed. [Results] UCP2 gene mutation difference was significant in both groups (P﹤0.05), yet PPAR- γ2 gene polymorphism and combined effects between the two variations were not significantly different (P﹥0.05). [Conclu- sion] The Ala55Val variation in UCP2 may be an alternative gene of simple obesity but the PPAR-γ2 gene Plo12Ala variation may not be considered as a risk factor of obesity in Chengdu region and without combined effects of the two variations.
