摘 要:【目的】观察C57BL/6哮喘小鼠肾组织瞬时感受器电位M6离子通道(TRPM6)mRNA的表达,探讨哮喘低镁血症的可能原因。【方法】健康4~6周龄清洁级雌性C57BL/6小鼠48只,体质量(12±2)g,随机数字表法分为哮喘组和对照组,每组各24只。哮喘组应用卵蛋白(OVA)建立哮喘小鼠模型。第1天(1d)、21d、34d天每组分别随机抽取8只检测血浆Mg2+、红细胞内Mg2+、肾组织TRPM6mRNA的表达。【结果】1d血浆Mg2+、红细胞内Mg2+、肾组织TRPM6mRNA表达水平哮喘组与对照组之间差异无显著性:[(0.85±0.07vs.0.89±0.12)mmol/L、(2.48±0.14vs.2.49±0.07)mmol/L、0.51±0.08vs.0.49±0.06,P均>0.05];21d哮喘组血浆Mg2+、红细胞内Mg2+、肾组织TRPM6mRNA表达水平均显著低于对照组:[(0.84±0.09vs.0.95±0.07)mmol/L、(2.39±0.14vs.2.44±0.09)mmol/L、0.32±0.06vs.0.52±0.05,P均<0.05];34d哮喘组血浆Mg2+、红细胞内Mg2+、肾组织TRPM6mRNA表达水平亦显著低于对照组:[(0.67±0.10vs.0.94±0.10)mmol/L、(2.17±0.08vs.2.43±0.08)mmol/L、0.24±0.05vs.0.53±0.06,P均<0.05]。肾组织TRPM6mRNA表达水平与血浆Mg2+浓度呈正相关(r=0.630,P<0.001);肾组织TRPM6mRNA表达水平与红细胞内Mg2+浓度呈正相关(r=0.715,P<0.001)。【结论】肾组织TRPM6mRNA的低表达可能是导致C57BL/6哮喘小鼠低镁血症的原因。[Objective] To observe the expression of transient receptor potential melastatin 6 (TRPM6) mRNA in renal tissues of C57BL/6 asthmatic mice and to explore the possible mechanism of hypomagnesemia in asthma. [Methods] Forty-eight healthy C57BL/6 mice, which were 4-6 w old, female, body mass (12 ± 2)g, were randomly divided into asthmatic group and control group with 24 mice in each group. The asthmatic group were sensitized and challenged by ovalbumin. Eight mice were randomly taken out from each group at I d, 21 d, 34d to detect plasma Mg^2+ intracellular Mg^2+and expression of TRPM6 mRNA in renal tissues. [ Results ] There were no significant differences in plasma Mg^2+ intraeellular Mg^2+ and TRPM6 mRNA expression of renal tissues between asthmatic group and control group at 1 d [(0.85 ± 0.07) vs. (0.89 ± 0.12) retool/L, (2.48 ± 0.14) vs. (2.49 ± 0.07) retool/L, 0.51 ± 0.08 vs. 0.49 ± 0.06, P 〉 0.05, respectively); plasma Mg^2+and intracellular Mg^2+ as well as TRPM6 mRNA of renal tissues in asthmatic group were significantly lower than that of control group at 21 d [ (0.84 ± 0.09) vs. (0.95 ± 0.07) mmol/L, (2.39 ± 0.14) vs. (2.44 ± 0.09) retool/L, 0.32 ± 0.06 vs. 0.52 ± 0.05, P 〈 0.05, respectively); plasma Mg^2+and intracellular Mg^2+as well as TRPM6 mRNA of renal tissues in asthmatic group were significantly lower than that of control group at 34 d [(0.67 ± 0.10) vs. (0.94 ± 0.10) mmol/L, (2.17 ± 0.08) vs. (2.43 ± 0.08) mmol/L, 0.24 ± 0.05 vs. 0.53 ±0.06, P 〈 0.05, respectively). Both plasma Mg^2+ and intraeellular Mg^2+ were positively correlated with TRPM6 mRNA expression in the renal tissues (r = 0.630, 0.715; P 〈 0.001, respectively). [Conclusion] Downregulation of TRPM6 mRNA in the renal tissues may provide a molecular mechanism for hypomagnesemia in C57BL/6 asthmatic mice.
