摘 要:目的:为临床周围神经损伤的移植治疗提供生物化的神经移植替代物。方法:将骨髓神经组织定向干细胞(NTC-SCs)种植于犬坐骨神经脱细胞神经支架,构建组织工程化神经,桥接缺损的大鼠坐骨神经,检测所构建生物化组织工程神经中骨髓NTCSCs的增殖、分化及神经性基因表达,观察该神经移植到体内后的细胞增殖、分化及对神经缺损功能的修复。结果:由骨髓NTCSCs和异种脱细胞神经支架构建的生物化组织工程神经物理性状相似于神经移植体,骨髓NTCSCs在支架上增殖、迁移,并表达神经性基因及蛋白。该组织工程神经植入体内后,骨髓NTCSCs分化为少量神经元特异性烯醇化酶(NSE)阳性细胞及大量S-100阳性细胞,并可修复缺损的坐骨神经功能。结论:骨髓NTCSCs和异种脱细胞神经支架构建的生物化组织工程神经相似于神经移植体,可应用于周围神经损伤修复中。Objective: To explore the feasibility of tissue engineered nerve for repairing the injured peripheral nerve, and to provide the experimental basis for clinical translation. Methods: The decellularized scaffolds prepared by using lyolecithin, DNase and RNase to treat the canine sciatic nerves were compounded with bone marrow derived neural tissue-committed stem ceils via microinjection to creat the tissue engineered nerve, and thereby bridge the mouse sciatic nerve gap. Proliferation, differentiation and expressions of neural specific genes of neural tissue-committed stem cells in vitro and in vivo were detected, and the recovery of nerve function was judged. Results: The creation of the engineered nerve which was reconstructed by tissue-committed stem cells and decellularized scaffold was similar to the homograft in physical properties. Migaration, proliferation and expression of neural specific markers both in mRNA and protein levels of neural tissue-commited stem cells seeding into decellurized scaffold were detectable in vitro. After implantation of engineered nerve into the sciatic nerve gap, a few neural tissue-committed stem cells were stained against neuron specific enclose (NSE), most cells were S- 100 positive, and the nerve function was restored. Conclusion: Tissue engineered nerve created hy neural tissue-committed stem ceils and decellurized scaffold have a potential to repair the injured peripheral nerve due to its similarity with homograft.
