摘  要:目的探讨血管紧张素II(AngII)对人脐动脉平滑肌细胞妊娠相关蛋白-A(PAPP-A)和胰岛素样生长因子-1(IGF-1)mRNA表达的影响。方法(1)应用10-5mol/LAngII与人脐动脉平滑肌细胞及在Ox-LDL中刺激过的人脐动脉平滑肌细胞共同培养0、6、12、24、36、48h后收集细胞。(2)应用不同浓度的AngII(10-7、10-6、10-5、10-4mol/L)与上述两种细胞共同培养24h后收集细胞。(3)应用RT-PCR方法检测细胞内PAPP-A、IGF-1mRNA的表达量。结果在浓度为10-5mol/L的AngII作用下,PAPP-A、IGF-1表达量在12h时开始升高(P<0.05),24h左右达高峰(P<0.01),而后表达量无进一步上升(P>0.05)。在不同浓度AngII刺激下,PAPP-A、IGF-1表达量随着浓度的升高而增加(P<0.05),10-5mol/L时达高峰(P<0.01),进一步加大浓度表达量无进一步升高(P>0.05)。Ox-LDL刺激过的细胞不同时间点以及不同AngII浓度下表达量明显高于同一时间及同一浓度时未刺激细胞的表达量(P<0.05)。结论在一定的作用时间和浓度范围内,AngII呈浓度和时间依赖性升高PAPP-A、IGF-1,从而影响动脉粥样硬化斑块的稳定性而导致急性冠脉综合症的形成,Ox-LDL可以促进这种作用。Objective To study the effect of angiotensin Ⅱ (Ang Ⅱ) on pregnancy-associated plasma protein A (PAPP-A) and insulin-like growth factor 1 (IGF-1) mRNA expressions in human umbilical artery smooth muscle cells (HUVSMCs). Methods In the presence or absence ofOx-LDL, HUVSMCs were cultured with Ang Ⅱ of 10^-5 mol/L for 0, 6, 12, 24, 36, and 48 h, or with Ang Ⅱ at the concentrations of 10^-7, 10^-6, 10^-5, and 10^-4mol/L for 24 h, after which the cells were theia collected to detect PAPP-A and IGF-1 mRNA expressions in the cells using RT-PCR. Results At the concentration of 10^-5 mol/L, Ang Ⅱ showed a time-dependent effect in inducing PAPP-A and IGF-1 mRNA expressions, which began to increase at 12 h of culture and reaching the highest level at 24 h. Ang II also dose-dependently induced PAPP-A and IGF-1 mRNA expressions, and 10.5 mol/L Ang Ⅱ induced the highest expression levels of the two genes. Ox-LDL exposure significantly further increased the expression levels of PAPP-A and IGF-1 mRNA in the cells regardless of the Ang Ⅱ concentration or duration for cell treatment (P〈0.05). Conclusion Ang Ⅱ can time- and dose-dependently induces PAPP-A and IGF-1 mRNA expression in HUVSMCs and is responsible for inducing platelet activity and inflammatory reaction in acute coronary syndromes, and the effects orang II can be enhanced by. Ox-LDL.
