自体角膜缘干细胞及羊膜对翼状胬肉TNF-α、IL-6表达的影响

摘　　要：目的研究自体角膜缘干细胞及羊膜对翼状胬肉肿瘤坏死因子α(tumor necrosis factor α,TNF-α)、白细胞介素6(interleukin IL-6)表达的影响。方法取门诊患者18例(18眼)翼状胬肉组织,随机分为3组(A组为单纯翼状胬肉组、B组为保存羊膜与翼状胬肉混合组、C组为自体角膜缘干细胞组织与翼状胬肉混合组),培养24小时后取各组翼状胬肉分别制备组织上清液,ELISA法检测TNF-α和IL-6含量。结果与A组比较,B组的TNF-α、IL-6含量均显著降低,差异有统计学意义(P<0.05);C组仅有TNF-α含量显著降低,差异有统计学意义(P<0.05),而IL-6含量变化不明显,差异无统计学意义(P>0.05)。结论保存羊膜可降低翼状胬肉TNF-α、IL-6的表达;自体角膜缘干细胞仅降低翼状胬肉TNF-α的表达,而对翼状胬肉IL-6的表达无影响。OBJECTIVE To study the effect of tumor necrosis factor α（TNF-α） and interleukin 6 （IL-6） in corneal stem cell transplantation or amniotic membrane transplantation in pterygia. METHODS Eighteen patients＇ （18 eyes） pterygia tissues were obtained and were randomly divided into the following 3 groups： group A was the pure pterygia group, group B was the conservative amnia with pterygia tissue group, group C was the corneal stem cell with pterygia tissue group. After being cultured for 24 hours, tissue supernatant of each group was tested for the content of TNF-α and IL-6 by enzyme-linked immunosorbent assay （Elisa technique）. RESULTS Compared with group A, the content of TNF-α and IL-6 in group B was significantly reduced （P〈0.05）. The content of TNF-α in group C was significantly reduced （P〈0.05）, but the content of IL-6 didn＇t changed much （P〉0.05）. CONCLUSIONS Conservative amnia could reduce the expression of TNF-α and IL-6 in pterygia;the corneal stem cell could reduce the expression of TNF-α, but had no effect on the expression of IL-6 in pterygia.
