积雪草对2型糖尿病大鼠血浆P-选择素、D-二聚体水平的影响

摘　　要：目的观察积雪草对2型糖尿病大鼠血浆P-选择素(Ps)、D-二聚体(D-D)水平的影响。方法采用尾静脉注射链脲佐菌素并高糖高脂饲料喂养诱导2型糖尿病大鼠模型,造模成功后2型糖尿病大鼠随机分为模型组(n=10)、二甲双胍组(n=11)、积雪草中剂量组(n=11)和积雪草高剂量组(n=11),以正常鼠为对照组(n=10)。治疗8周后采用酶联免疫吸附分析(ELISA)法检测各组血浆Ps、D-D的水平。结果模型组Ps、D-D水平较正常对照组明显增高(P<0.01),药物治疗后,积雪草组与二甲双胍组大鼠Ps、D-D水平均明显下降(P<0.05,P<0.01)。其中,积雪草高剂量组D-D水平较二甲双胍组下降更明显(P<0.05)。结论积雪草能明显降低糖尿病大鼠血浆Ps、D-D水平,提示积雪草具有减少血小板活化,改善凝血纤溶功能的作用,进而有助于预防和延缓糖尿病血管病变的发生发展。Objective To observe the effects of the herb Jixuecao on the levels of P-selectin and D-Dimer in rats with type 2 diabetes. Methods The rat model of type 2 diabetes were induced by an intravenous injection of streptozotocin from caudal vein and a forage feeding with high glucose and high grease. Then the type 2 diabetes rats were randomly divided into 4 groups ： model group（ n = 10） , Metformin group（ n = 11 ） ,Jixueeao middle-dose group（ n = 11 ） and Jixuecao highdose group （ n = 11 ）. There was also 10 animals in the normal control group. After 8 weeks＇ therapy, the levels of P-selectin and D-Dimer in blood plasma between the five groups were detected by enzyme linked immunosorbent assay（ELISA）. Resuits The levels of P-selectin and D-Dimer in the model group were significantly higher than that in the normal control group （P 〈 0.01 ）, and the levels in the Metformin group and the Jixuecao groups were statistically decreased after the medicine treatments, especialliy in the Jixueeao high-dose group compared with the Metformin group. Conclusion The herb Jixuecao lowered the levels of P-selectin and D-Dimer in blood plasma in rats with type 2 diabetes, indicating that Jixuecao could decrease the activation of platelet and improve the fuction of blood clotting and fibrinolysis, which conduces to prevent and postpone the occurrence and development of diabetes vasculopathy.
