摘  要:目的探讨变应性鼻炎(allergic rhinitis,AR)小鼠鼻黏膜核因子-κB(nuclear factor kappat B,NF-κB)p65和糖皮质激素受体(glucocorticoid receptor,GR)的表达,进一步了解NF—κB p65和GR在AR发病机制中的作用。方法清洁级C57BL6/J小鼠20只,随机分为AR组和对照组,每组10只。AR组用10%卵清蛋白(ovalbumin,OVA)200μl腹腔注射,6%OVA鼻腔局部激发建立AR模型。取鼻面骨,石蜡包埋,连续切片,行HE染色。分别运用免疫组化SABC法和SP法染色,观察鼻黏膜中NF-κB p65和GR的表达情况。结果AR组中10只动物均建模成功,鼻部AR典型症状明显;对照组无明显变化。①形态学改变:对照组鼻黏膜无明显炎症改变;AR组黏膜则显示不同程度水肿,黏膜下嗜酸粒细胞(eosinophils,Eos)、淋巴细胞和单核细胞浸润。②黏膜NF—κB p65表达:NF-κB p65主要表达于上皮细胞和腺上皮细胞的胞浆,偶见胞核。AR组中上皮细胞染色平均光密度值(0.40±0.12)强于对照组(0.23±0.05;P〈0.05)。③GR表达:2组中均有GR表达,主要表达于鼻黏膜上皮、腺上皮及血管内皮细胞的胞核或胞质。AR组中GR的平均光密度值(0.20±0.04)低于对照组(0.36±0.11;P〈0.05)。结论 NF-±κB p65在AR黏膜中表达增强,而GR的表达下降,提示两者的失衡在AR的发病中发挥重要作用。Objective To investigate the influence of allergic rhinifis on the expression of nuclear factor- κB (NF-B) p65 and glucocorticoid receptor(GR) in nasal mucosa, Methods 20 mice of C57BL6/J were randomly divided into 2 groups equally: control group and allergic rhinitis (AR) model group. AR model group were sensitized by intraperitoneal injection 200μlOVA (10%) on days 1 to 9, and exposed to OVA (6%) intranasally on days 10 to 17. SO as to induce allergic inflammation. PBS was used in control group in the same way. The heads were embedded with paraffin and serial sections were made and stained with hematoxlin-eosin for histological analysis. The expressions ofNF- κB p65 and GR after immunohistochemical staining by SABC and SP respectively were observed. Results AR models were successfully established in 10 mice from AR model group, ①Pathomorphology changes of nasal mucosa: In AR model group, the remarkable symptoms of infiltrations of inflammatory cells such as eosinophils, mononuclear ceils and lymphocytes, were fotmd in nasal mucosa. In control group, no abnormality was observed. ②Expression ofNF- κB p65: In AR model group, Plasma and nucleal of epithelial and glandulal cellsdisplayed a more strongly positive staining reaction to NF- κB p65. The average optical of NF- κB p65 (0. 40± 0.12) is higher than in control group (0. 23 ±0. 05. P〈0.05. ③Expression of GR: both groups, plasma and nudear of epithelial, vascular endothelial and glandular cells displayed a positive staining reaction to GR, but the average optical in control group (0. 36 ±0. 11)was more strong, than in AR model group (0. 20 ±0. 04 P〈0. 05). Conclusions Our results suggested that the occurrence of allergic rhinitis inight,be related with the activation of NF- κB p65 and down regulation of expression of GR The disequilibrium of NF- κB p65 / GR might play an important role in the development of allergic rhinitis.
