摘 要:目的探讨经Cad-Ⅱ(OB-Cadherin,成骨细胞特异性钙粘蛋白)基因转染的人骨髓间充质干细胞(humanmarrow mesenchymal stem cells,hMSCs)在骨基质材料上的粘附性和增殖情况。方法将脂质体介导的Cad-ⅡcDNA转染体外分离培养的hMSCs,再将已转染和未转染的hMSCs分为实验组和对照组,分别与异种冻干脱钙骨基质材料复合体外构建组织工程骨,通过相差显微镜、扫描电镜及MTT检测对比观察两组细胞在材料上的黏附和增殖情况。结果2组细胞均能在骨基质材料上粘附、伸展、增殖,其中增殖情况无明显差异(P>0.05),但Cad-Ⅱ基因转染组的上架细胞数和上架率(84.5%),显著高于未转染组(71.7%),差异显著(P<0.05)。结论Cad-Ⅱ基因转染能提高种子细胞在支架材料上的粘附性,但对细胞增殖无影响。Objective To study the effect of Cad-Ⅱ gene (OB-Cadherin) transfection on the adhesion and proliferation of human marrow mesenchymal stem cells (hMSCs) cultured on xenogeneic demineralized bone matrix. Methods The Cad-Ⅱ gene was transfected into hMSCs, and those without transfection served as control group. Then they were seeded onto the xenogeneic demineralized bone matrix respectively and cocultured in vitro. The densities of seeded cells and adhesion rate was measured, the adhesion with bone matrix complex was also observed by contrast microscopy and electron scanning microscopy. And the cell proliferation was measured with MTT method. Results The 2 groups of hMSCs adhered to the surface and inner walls of bone matrix, and spread extensively, with no significant difference in cell proliferation between them. But in the transfection group there were more adhesion cells on the scaffolds, and the adhesion rate ( 84.5 % ) was significantly higher than that in control group (71.7%, P 〈 0.05). Conclusion The Cad-Ⅱ gene transfection may enhance the seeded cells adhesion to scaffolds , but has no obvious effect on cell proliferation.
