摘 要:目的研究EPO对人CD34+脐血干细胞表面CD26表达的影响。方法应用流式细胞仪法检测经MACS-CD34+免疫磁珠分选并采用EPO处理后培养的人CD34+脐血干细胞,通过与酶底物反应检测人CD34+脐血造血干细胞表面CD26的表达情况。结果与对照组相比,经EPO 100 U/ml和EPO 200 U/ml 18h培养后的CD34+脐血造血干细胞群中CD26+细胞比例明显上升,其酶活性亦明显升高。结论EPO可上调CD34+脐血干细胞表面CD26的表达并增强其生物学功能。Objective To observe short-term erythropoietin (EPO) treatment on the expression of CD26, dipeptidyl-peptidase IV, in the CD34^+ human cord blood hematopoietic ceils in vitro. Methods The CD34^+ cells were sorted from human cord blood by magnetic activated cell sorting (MACS) technique, and subjected to recombinant human EPO treatment at a dose of 50, 100 and 200 U/ml. We detected CD26 expression of the treated and non-treated cells by flow cytometry and CD26 activity by responding with chromogenic CD26 substrate Gly-Pro-p-nitoanilide. Results CD26 expression and activity increased significantly in EPO 100 and 200 U/ml treatment group. Conclusion Short-term EPO treatment in vitro upregulates CD26 expression and activity in the CD34^+ human cord blood hematopoietic cells, and enhances its biological function.
