摘 要:目的初步研究S180、H-22肿瘤细胞对超声杀伤作用的敏感性比较及其生物学机制。 方法频率2.17MHz,强度3w/cm^2的聚焦超声处理细胞后,分别检测其存活率、FD500阳性细胞率、形态变化、LDH释放量、活性氧产生以及脂质过氧化水平。 结果S180与H-22细胞发生不同程度的膜通透性和表面超微结构改变、LDH释放量增加、胞内活性氧和脂质过氧化水平增高。 结论超声对2种细胞均产生损伤效应,S180细胞对超声较H-22细胞更为敏感,其差异机制可能归咎于细胞间不同的化学组成和分子结构。Objective This assay intends to investigate the differences of sensitivities between S180 and H-22 tumor cells to focused ultrasound and the potential mechanisms. Methods After exposed to focused ultrasound of 2.17MHz at 3W/cm^2 , viabilities of tumor cells, FITC dextron positive cells, Morphological changes, Lactic Dehy drogenase (LDH) release, Reactive oxygen species (ROS), Lipid peroxidation level was examnied. Results Either cell lines exhibit different level of damage decreased viabilities, morphological changes, increased FITC dextron positive cells, higher oxidative stress and more amounts of ROS. Conclusions Both cell lines were damaged by focused ultrasound, while the S180 cell represents deeper damage than the H-22 cells. The mechanism of this difference may be ascribed to the different chemical compositions, molecular structure of the cell.
