摘 要:目的观察宝灵膏对蒙古沙鼠脑缺血再灌注损伤脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)、B细胞淋巴瘤/白血病-2(Bcl-2)表达的影响及是否存在剂量依赖性。方法采用双侧颈总动脉阻断的方法建立沙鼠脑缺血再灌注损伤模型。沙鼠随机分为假手术组、模型组、宝灵膏高剂量治疗组、宝灵膏中剂量治疗组、宝灵膏低剂量治疗组。采用比色法测量脑组织中SOD、MDA的含量,采用免疫组织化学法检测Bcl-2的阳性细胞数。结果宝灵膏各剂量治疗组SOD、MDA、Bcl-2与模型组差异明显,宝灵膏高、中剂量组SOD、MDA、Bcl-2与低剂量组亦有明显差异,与假手术组相近。结论宝灵膏能促进在沙鼠脑缺血再灌注损伤中对抗氧自由基损伤的SOD的表达、降低MDA的表达、上调Bcl-2的表达,从而起到对抗氧自由基对脑组织的损伤及抑制脑神经细胞的凋亡和坏死的作用,并且其作用具有剂量依赖性。Objective: To observe the effects of bowring paste(BP)on expression of superoxide dismutase(SOD), malondialdehyde (MDA), B cell lymphoma/leukemia - 2 (Bcl - 2 ) after cerebral isehemia reperfusion injury (IRI) in mongolian gerbil and the influence whether show of dose - depended manner. Methods: Bilateral carotid artery occlusion method was used to establish gerbil cerebral ischemia - reperfusion injury model. The gerbils were randomly divided into sham group, model group, bowring paste highdose treatment group, howring paste medium- dose treatment group, bowring paste low - dose treatment group. The SOD, MDA of brain tissue were measured by col- orimetry and the Bel - 2 positive cells were measured by immunohistochemistry. Results: The SOD MDA and Bcl - 2 of three bowring paste treatment groups had obvious difference with these of model group. The SOD, MDA and Bcl - 2 of bowring paste high, medium - dose treatment group also had obvious difference with these of bowring paste low - dose treatment group, and resembled sham group. Conclusion: Bowring paste can promote the expression of SOD in the gerbil cerebral ischemia reperfusion injury, reduce the expression of MDA, and increase the expression of Bcl - 2, thus play a very good pair of free radical oxidation of brain damage and inhibit cells in the role of apoptosis and necrosis,and its role in dose- dependent.
