摘  要:采用随机扩增多态DNA(RAPD)技术对取自海南(HN)、浙江(ZJ)、江苏(JS)、河北(HB)等近海海域的4个地理群体银鲳进行遗传多样性分析。从20个随机引物中筛选出10个,利用POPGEN Version 1.31软件对多态位点比例和遗传距离进行统计分析。共检测出总位点数56个,其中多态位点有39个,多态位点比例为69.64%。4个海域银鲳群体的Shannon多样性指数分别为0.1232—0.1587,Nei基因多样性指数分别为0.0776—0.1004。JS群体与HN群体的遗传距离最远,为0.3216;JS群体与HB群体的遗传距离最近,为0.2316。结果表明,RAPD技术具有较高的灵敏性和多态性,是分析评估银鲳遗传多样性较为适宜的分子标记之一;JS群体保持着最高的遗传多样性,而HN群体多态性比例最低、ZJ群体Shannon多样性指数最低,应采取一些有效的保护措施,以避免HN群体遗传多样性的进一步丧失。The present paper deals with the preliminary study on the genetic diversity of four geographic populations of silver pomfret ( Pampus argenteus ) by means of random amplified polymorphie DNA (RAPD) technique. Samples were respectively caught from the sea areas near the coast of Xinbudao Is. of Hainan ( HN), Zhoushan Is. of Zhejiang ( Z J), Lianyungang of Jiangsu (JS) and Qinhuangdao of Hebei (HB) province. It was used POPGEN Version 1.31 software on the proportion of polymorphic locis about genetic distance and statistical analysis by 10 primers selected from 20 random primers. It had been detected in a total of 56 sites and 39 polymorphic sites. The proportion of polymorphic sites was 69.64 %. The Shannon diversity index of the 4 geographic populations were 0. 123 2 - 0. 158 7 ; Nei genetic diversity index was 0. 077 6 - 0. 100 4.The distance between population JS and HN was the farthest (0. 3216). However, the distance between population JS and HB was the nearest (0. 2316). The results showed that RAPD technique, with its high sensitivity and polymorphism, was suitable for the study on genetic diversity of P. argenteus populatons; JS population still maintained the highest genetic polymorphic sites and Shannon diversity index. measures to avoid further loss of genetic diversity diversity but HN population kept the lowest proportion of It could be inferred that we should take some effective of HN population.
