异叶败酱中三萜化合物常春藤皂苷元对人早幼粒白血病细胞HL-60的增殖抑制、周期阻滞及凋亡诱导作用

摘　　要：利用倒置显微镜观察法、台盼蓝排染法、Hoechst33258荧光染色法、DNAladder电泳以及流式细胞分析技术,检测了从甘肃产异叶败酱(Patrinia heterophyllaBunge.)中分离得到的三萜化合物常春藤皂苷元(HG)对人早幼粒白血病细胞HL-60的增殖抑制、周期阻滞及凋亡诱导作用.结果表明,常春藤皂苷元在较低浓度(10～40μmol·L-1)条件下,对HL-60细胞增殖具有良好的抑制作用;在较高浓度(40～50μmol·L-1)条件下,对HL-60细胞具有致死作用;同时,常春藤皂苷元对HL-60细胞的G1期阻滞和凋亡诱导作用极可能是其实现增殖抑制和致死作用的主要途径.The effects of Hederagenin（HG）, a triterpnoid from Patrinia heterophylla Bunge. , on the proliferation inhibition, cell cycle arrest and apoptosis of Human promyelocytic leukemia HL-60 cells are measured by morphological observation, typanblue exclusion method, Hoechst 33258 stain assay, DNA ladder and flow cytometry respectively. The results show that Hederagenin can inhibit the proliferation of HL-60 at low concentrations（10-40 μmol·L^-1） while make the cells death at the high concentrations（40 -50 μmol·L^-1）; HG can make proliferation inhibition and cell death on HL-60 cells through G1 phase arrest and apoptosis.
