邻苯二甲酸二丁酯对小鼠骨髓微核和精子的影响

摘　　要：目的采用小鼠骨髓微核试验和F1代雄性大鼠的生殖毒性实验研究邻苯二甲酸二丁酯(DBP)的遗传毒性和生殖毒性,为综合评价其安全性提供科学依据。方法昆明小鼠分为33、330、3300 mg/kg DBP剂量组,经灌胃染毒,按常规方法进行小鼠骨髓微核试验。选择妊娠期和哺乳期SD雌性大鼠,灌胃染毒,DBP剂量分别为0、125、250、500 mg/kg,计数F1代雄性大鼠的附睾尾精子数、精子活力、精子畸形率。结果DBP在3 300 mg/kg时能引起微核率的升高(P<0.01)。中、高剂量组(250、500 mg/kg)DBP可引起F1代仔鼠精子总数、精子活力明显降低(P<0.01),精子畸形率升高(P<0.05)。结论DBP具有一定的遗传毒性和生殖毒性。Objective The genotoxicity of dibutyl phthalate （DBP）was examined using the mice bone -narrow micronucleus test and the reproductive toxicity was examined using reproductive system of F1 generation male rats toxicity test. Methods Mieronucleus test： Kuming mice were divided into 3 groups at the dibutyl phthalate dosage of 33,330,3 300 mg/kg, and the bone - narrow micronueleus test was undertaken according to the routine method. Reproductive toxicity test： the SD pregnant rats were treated with different dose of dibutyl phthalate （125,250 and 500 mg/kg） intragastric administration during the gestation and lactation period. The quantity of sperm in cauda epididymidis ,the motility of sperm and the abnormality rate of sperm of F1 generation were counted. Results Compared with those in the negative control group, the rates of micronucleus of DBP significantly increased only at dosage of 3 300 mg/kg treatment group （ P 〈 0.01 ）. Meanwhile ,the quantity of sperm in cauda epididymidis and the motility of sperm at dosage of 250 and 500 mg/kg groups were significantly decreased（ P 〈 0.01 ）compared with those in the negative control group, the abnormality rate was significantly increased（ P 〈 0.05 ）. Conclusion The ：esearch suggested that there were some genotoxicity and reproductive toxicity risks of DBP.
