摘 要:目的:探讨金葡菌肠毒素B(staphylococcal enterotoxin B,SEB)对支气管哮喘豚鼠模型支气管肺泡液高迁移率族蛋白B1(high mobility group protein B1,HMGB1)及Th1/Th2细胞因子水平的影响。方法:18只豚鼠随机分为对照组6只、支气管哮喘豚鼠模型组6只和SEB组6只,模型组用卵白蛋白(ovalbumin,OVA)建立豚鼠哮喘模型,SEB组在OVA致敏前10 d于腹腔注射SEB 1 ml(10 mg/L),对照组注射等量生理盐水。支气管哮喘诱发后2 d,收集支气管肺泡灌洗液进行白细胞和嗜酸性粒细胞计数,WB法测定HMGB1水平,ELISA法检测细胞因子IFN-γ和IL-4水平。结果:模型组肺泡灌洗液白细胞数、嗜酸性粒细胞数及比例、HMGB1水平和IL-4浓度显著升高,IFN-γ浓度显著降低。SEB组白细胞数、嗜酸性粒细胞数量及比例、HMGB1水平和IL-4浓度显著低于模型组,IFN-γ浓度高于模型组。结论:SEB能减少哮喘豚鼠支气管肺泡灌洗液中HMGB1水平及嗜酸性粒细胞数量及比例,其机制可能与Th1/Th2细胞因子比值调节有关。Objective: To study the effects of staphylococcal enterotoxin B (SEB) on high mobility group protean B1 ( HMGB1 ) and the IFN - γ/IL - 4 balance in bronchoalveolar lavage fluid (BALF) from ovalbumin ( OVA ) - challenged and sensitized guinea pig. Methods : Eighteen guinea pigs were divided into the control group, the asthma model group ( the model group) and the SEB group at random. Each group had 6 guinea pigs. The pigg asthma in the model group was induced by OVA. In the SEB group, 10 μg SEB was injected intraperitoneally 10 days before sensitization with OVA. Two days after the last challenge, the number of WBC and eosinophils in BALF was counted, the concentration of HMGBI in BALF was detected by WB technique and the concentrations of IFN - γand IL - 4 were detected by ELISA technique. Results : The number of WBC,eosinophils,eosinophils/WBC,HMGB1 and IL - 4 level of BALF in the model group were higher than those in the control group except the IFN - 7 level in BALF of the model group decreased significantly. Compared with the model group .the number of WBC. eosinophils, eosinophils/ WBC, HMGB1 and IL - 4 level of BALF were lower while the IFN - γ level of BALF was significantly higher in the SEB group. Conclusion:SEB can decrease the amount of WBC, eosinophils and HMGBI level in BALF in guinea pigs with asthma, which may be related to the accommodation of IFN -γ/IL - 4 in lung tissues.
