摘  要:目的:探讨有氧运动对高脂饮食大鼠血脂及骨骼肌过氧化物酶体增殖物激活受体-α(PPARα)、三磷酸腺苷结合盒转运体A(lABCA1)、载脂蛋白AI(ApoAI)基因表达的影响。方法:将40只SD雄性大鼠分为4组:正常饮食组(N)、正常运动组(NE)、高脂饮食组(H)和高脂运动组(HE)。H组和HE组大鼠采用高脂饲料喂养。NE组和HE组大鼠进行跑台运动,速度为15m/min,每天60min,每周5d,持续8周。实验结束后,采用酶法、酶修饰法和清除法测试各组大鼠血清甘油三酯(TG)、胆固醇(TC)、低密度脂蛋白(LDL-C)与高密度脂蛋白(HDL-C);采用实时定量PCR测定骨骼肌PPARα、ABCA1、ApoAI mRNA表达。结果:(1)与N组比较,H组TG、TC、LDL-C水平显著上升,而HDL-C水平显著下降;HE组TG含量显著低于H组,HDL-C水平显著高于H组。HE组与H组相比,血清TC、LDL-C水平无显著性差异。(2)NE组大鼠PPARα、ABCA1、ApoAI mRNA的表达较N组显著增加;H组PPARα、ABCA1、ApoAI mRNA表达与N组相比显著降低;HE组ABCA1、ApoAI mRNA的表达较H组显著提高,而PPARα mRNA的表达与H组比较无显著差异。结论:(1)高脂饮食可以导致血脂水平异常,并使骨骼肌PPARα、ABCA1、ApoAI mRNA表达显著下降;(2)运动可以一定程度改善高脂饮食导致的血脂异常;运动可上调高脂饮食大鼠骨骼肌ABCA1和ApoAI mRNA表达。PPARα在该通路中可能不是唯一的调节机制。Objective To discuss the effects of 8-week-exercise and high-fat diet on the changes of blood lipid, peroxisome proliferators activated receptor α(PPARα), ATP-binding cassette transporter A1 (ABCA1) and apolipoprotein AI(ApoAI) mRNA in SD rats. Methods 40 male SD rats were randomly divided into 4 groups: normal diet group(Group N), normal diet with exercise group(Group NE), high-fat diet group(Group H) and high-fat diet with exercise group(Group HE). The rats in group N and NE were fed with normal diet, and those in group H and HE were fed with high-fat diet. The exercise program were conducted on treadmill in medium intensity (15m/min, 60min/d 5days/week for 8 weeks). TG,TC,LDL-C and HDL-C in serum were measured by enzymatic and enzymatic modified method. The expression of PPARα, ABCA1, ApoAI mRNA were measured by Real-Time PCR. Results (1)The TG, TC,LDL-C of group H increased and HDL-C of group H decreased significantly compared with group N. Compared with the group H ,TG of group HE were significantly lower and HDL-C were significantly higher. No significant difference of TC and LDL-C had been found between group HE and H.(2)Compared with group N,the express of PPARα,ABCA1 and ApoAI mRNA significantly increased in group NE and significantly decreased in group H; PPARα mRNA of group HE showed no significant difference,but ApoAI mRNA were significantly higher than group H. Conclusions (1)High-fat diet could induce abnormal blood lipid and increase the express of PPARα,ABCA1 and ApoAI mRNA in muscles; (2) Aerobic exercise could improve the high-fat diet caused abnormal blood lipids and upregulate the express of ABCAI ,ApoAI mRNA in rat muscles. The result suggested that PPARα might not be the only regulate mechanism of lipid metabolism.
