绵羊多胎主效基因Fec^B分子检测方法的建立与应用

摘　　要：Fec(BFec,fecundity;B,Booroola)是在布鲁拉美利奴(Booroola Merino)绵羊(Ovis aries)中发现的能增加排卵数和产羔数的一个常染色体突变基因,是在绵羊中识别出的第1个高繁殖力主效基因。建立了绵羊多胎主效基因FecB的PCR-RFLP和PCR-SSCP检测方法,对小尾寒羊、湖羊、中国美利奴、东弗里生、杜泊、萨福克、特克塞尔、多赛特、考力代、德国肉用美利奴、南非肉用美利奴以及杂种羊共6 647只绵羊的检测结果表明,两种检测方法均能准确判型,稳定可靠性好,而且两种方法的检测结果完全一致。检测结果表明小尾寒羊和湖羊携带FecB突变(A746G),中国美利奴、东弗里生、杜泊、萨福克、特克塞尔、多赛特、考力代、德国肉用美利奴和南非肉用美利奴绵羊都不携带该突变。The Booroola gene Fec^B（Fec, fecundity; B, Booroola） is the major gene for prolificacy firstly identified in sheep （Ovis aries）. The Fec^B locus is situated in the region of ovine chromosome 6 corresponding to the human chromosome 4q22-23 that contains the bone morphogenetic protein receptor IB （BMPR-IB） gene. A nonconservative substitution （Q249R） in the BMPR-IB coding sequence was fully associated with the hyperprolific phenotype of Booroola ewes. PCR-RFLP and PCR-SSCP methods for high prolificacy major gene Fec^B in sheep have been established in the present study. The detection results of 6 647 sheep including Small Tail Han, Hu, Chinese Merino, East Friesian, Dorper, Suffolk, Texel, Dorset, Corriedale, German Mutton Merino, South African Mutton Merino ahd crossbred sheep indicated that these two methods had the identical results and all displayed accurately genotyping, good stability and reliability, and showed that both Small Tail Han and Hu sheep carried the same Fec^B mutation （A746G） as found in Booroola Merino sheep. Chinese Merino, East Friesian, Dower, Suffolk, Texel, Dorset, Corriedale, German Mutton Merino and South African Mutton Merino sheep did not have Fec^B mutation.
