摘　　要：目的调查同一供体来源HIV-1病毒包膜序列变异情况,为进一步进行表型特征、病毒感染能力以及抗病毒免疫研究奠定基础。方法从6例同一来源HIV-1感染者外周血单个核细胞中提取DNA,设计跨病毒包膜的保守引物,进行PCR扩增,将扩增产物克隆至质粒载体,筛选阳性克隆进行序列测定。采用生物信息学方法对其进行系统发育特征以及编码氨基酸的变异性进行了分析。结果共获得6个有完整开放读码框的包膜克隆。在6个克隆中,包膜糖蛋白gp120内变异程度大的区域分别为信号肽区、可变区V1/V2区、恒定区C2区内近V3区部分、V4区和V5区。变异包括点突变、氨基酸插入或缺失等。系统树分析发现所获得病毒克隆与中国分离株BCNB03聚集成一束。结论构建并鉴定了6个HIV-1包膜克隆。同一来源的HIV包膜糖蛋白在不同受者体内发生了较大变异。Objective To investigate the variability of HIV-1 envelope （env） glycoprotein genes derived from the same donor. Methods Full-length env genes amplified from PBMC of six HIV-1 positive individuals from the same donor in Heilongjiang province in China using conserved region primers were cloned into a plasmid vector and sequenced. The analysis on the variability and the phylogenesis was done upon the env region. Results Six functional env clones from 6 infected individuals were obtained. The results showed that the most variable regions in the env glycoprotein were signal peptide region, variable region V1/V2, conserved region C2 neighboring to V3 region and variable region V4 and V5, respectively. The variations include site mutation, insertion and deletion of amino acids. Phylogenetic analysis indicates that all env genes isolated from Heilongjiang province were clustered with the isolate BCNB03 in China. Conclusion Six HIV-1 primary isolate envelope genes have been cloned from six HIV-1 positive individual in Heilongjiang province and sequenced. Bioinformatics analysis indicates that there is definite env variation among the recipient viruses derived from the same donor.
