肺炎支原体诱导A549细胞分泌肿瘤坏死因子-α和白细胞介素-8的相关性

摘　　要：目的:观察肺炎支原体(MP)诱导A549细胞产生肿瘤坏死因子-α(TNF-α)和白细胞介素-8(IL-8)的相关性,为MP肺炎的临床诊治提供依据。方法:体外培养A549细胞和MP,用感染复数为1:20的MP加入到A549细胞中共同孵育,采用双抗体夹心ELISA法检测不同刺激时间的MP作用A549细胞后分泌TNF-α和IL-8的含量。结果:MP能显著诱导A549细胞分泌TNF-α和IL-8(均P<0.01),经相关回归分析,其产生的水平与刺激时间具有依赖性(r=0.821,0.911,P<0.01),且二者之间具有量的正相关(r=0.942,P<0.01)。结论:MP诱导A549细胞产生的炎性细胞因子TNF-α和IL-8具有量的正相关性。Objective： To investigate the relation of cytokines tumor necrosis factor-α （TNF-α） and interleukin-8 （IL-8） secretions in A549 cells which were induced by Mycoplasma pneumoniae （MP）. Methods： A549 cells and MP were cultured in vitro. MP of multiplicity of infection （MOI） 1 ： 20 were co-cultured with A549 cells in vitro. Enzyme-linked immunosorbent assay （ELISA） was used to detect the contents of TNF-α and IL-8 in different time. Results： MP could significantly induce TNF-α and IL-8 secretions in A549 cells （P〈0.01, respectively）. With the correlation and regression analysis, the level of TNF-α or IL-8 was respectively depended on stimulation time （r=0. 821, P〈0.01 ; r=0. 911, P〈0.01）. And there was a positive correlation between the content of TNF a and IL-8（r=0. 942, P〈0.01）. Conclusion： There is a positive correlation between the cytokine TNF-α and IL-8 secretions induced by MPs in A549 cells.
