联合用药预防激素性股骨头坏死对骨细胞凋亡和Bcl-2表达的影响

摘  要:目的观察联合用药预防激素性股骨头骨细胞凋亡的可能性。方法30只兔随机分成三组:对照组同期肌注生理盐水;激素组是给予糖皮质激素肌注;治疗组在给予同剂量糖皮质激素同时,每日使用血塞通、脂必妥和阿仑膦酸钠;用末端原位标识(TUNEL)法及免疫组化技术分别检测骨细胞凋亡和bcl-2表达情况,电镜观察股骨头组织学变化。结果电镜下激素组骨细胞核体积缩小,染色质边聚和浓密多见于对照组和治疗组;激素组凋亡细胞比率显著高于治疗组,而其Bcl-2表达则较治疗组低。结论联合应用血塞通、脂必妥和阿仑膦酸钠能抑制兔激素性股骨头细胞凋亡。Objective To explore the effect of drugs preventing teroid-induced osteonecrosis of the femoral head on apoptosis and the expression of Bcl-2 protein of osteocytes. Methods Thirty rabbits were divided randomly into 3 groups: group A, guoup B and group C. Rabbits in group A were injected with normal saline; rabbits in group B were treated with glucocorticoid; rabbits in group C were treated with glucocorticoid as that in group B, and meanwhile, orally received the panax notoginseng saponins, zhibituo and alenduonate for 8 weeks. Histopathological changes , apoptosis and the expression of Bcl-2 protein of femoral head were observed. Results Under electron microscope, compared with group A and group C, group B showed agreat deal of necrotic osteocytes with pyknotie nucleus, condensed chromatin and increased electron dense; Apoptosis in group B was more than that in group C, while the expression of Bcl-2 protein in group B was less than that in group C. Conclusion The zhibituo, panax notoginseng saponins and alenduonate may inhibit apoptosis of hormonal necrosis of femoral head of rabbits.
