猪胸膜肺炎放线杆菌ApxⅡ-ELISA抗体检测试剂盒的研制与应用

摘　　要：根据胸膜肺炎放线杆菌ApxⅡ毒素的特点,建立了一种能够诊断猪传染性胸膜肺炎和进行免疫后效果评估的血清学方法ApxⅡ-ELISA。本研究将该诊断方法进一步研制成试剂盒,对其特异性、敏感性、重复性和稳定性进行了测试,与间接血凝试验试剂盒进行了对比研究,并用该试剂盒检测了来自中国、英国、丹麦、美国、加拿大和澳大利亚的临床猪血清1 453份。结果表明,ApxⅡ-ELISA试剂盒具有很高的特异性和敏感性;3批试剂盒的批内和批间的重复性良好(变异系数均<15%);试剂盒在2～8℃保存9个月,稳定性良好;ApxⅡ-ELISA试剂盒能够有效的诊断猪传染性胸膜肺炎,同时也可作为一种评价猪群免疫后免疫效果的有效方法。According to the characterization of Actinobacillus pleuropneumoniae （APP） ApxⅡ , we developed a diagnostic kit, whose specificity, sensitivity, repeatability and stability were evaluated and compared with the indirect haemagglutination assay （IHA）. A total of 1 453 clinical sera from China, UK, Denmark,USA,Canada and Australia were screened using our Apx Ⅱ-ELISA kit. The results showed that the Apx Ⅱ-ELISA kit has very high specificity. sensitivity and repeatability （the inter-and intrabatch imprecision （CV % ） 〈15 %） and stability （very stable at 4-8℃ for 9 months）. Compared with the indirect haemagglutination assay （IHA）, Apx Ⅱ-ELISA is more sensitive than IHA. Our data confirmed that the ApxⅡ -ELISA kit can be used to detect the antibodies of the toxin Apx Ⅱ and evaluate the effect of App vaccine.
