摘　　要：建立组织贴块法培养原代和传代肉鸡肺动脉平滑肌细胞(PASMC)。将1～10日龄雏鸡的肺动脉中膜切成小于1 mm2的组织块,用翻转干贴壁法进行贴块培养。采用形态学和抗α肌动蛋白抗体(α-actin)免疫组化染色法对所培养的细胞进行鉴定。倒置显微镜观察培养的细胞呈梭形,重叠生长,呈典型的"峰-谷"状。抗α-actin单克隆抗体免疫组化染色可见胞浆内有明显的细丝,呈现棕黄色,表明所培养的细胞为肺动脉平滑肌细胞。组织贴块法简便易行,可用于血管病理生理变化研究。To culture pulmonary artery smooth muscle cells of broiler with the adherent method of tissue explants,and to subculture them. Freshly dissected pulmonary artery media layer from 1 to 10 day old broilers was rinsed and cut into less then 1 mm × 1 mm pieces for culturing of pulmonary artery smooth cells by method of adher- ent method of tissue explants. Identifications of the cells were performed with the morphological feature and immunohistochemical stain using monoclonal antibody against smooth muscle alpha actin. The result showed that cells were typical form of vascular smooth cells under light microscope and immunohistochemical test. This method is simple and convenient. It provides ideal materials for vascular physiopathologic studies.
