中药芪红合剂含药血清对人外周血内皮祖细胞增殖的影响

摘　　要：目的观察中药芪红合剂含药血清对体外培养的人外周血内皮祖细胞(EPCs)增殖的影响。方法将12只雄性Wistar大鼠随机分为中药芪红合剂组(低剂量组、中剂量组、高剂量组)和对照组,制备含药血清和空白血清。采用密度梯度离心法分离获得人外周血单个核细胞,经激光共聚焦显微镜、流式细胞仪鉴定为EPCs。加入各实验血清组培养液培养EPCs,分别作用不同时间(24h、48h、72h)后,倒置相差显微镜下观察EPCs生长情况,噻唑蓝比色试验(MTT法)测定EPCs的吸光度值。结果与对照组比较,中药芪红合剂含药血清低、中、高剂量组EPCs增殖明显(P<0.05),且呈一定的量效、时效关系。在同一作用时间,低剂量芪红合剂含药血清干预后EPCs的增殖明显(P<0.05)。在同一剂量组中,芪红合剂含药血清干预后EPCs的增殖随时间推移逐渐增强,于72h达到高峰(P<0.05)。结论中药芪红合剂含药血清对体外培养的人外周血EPCs的增殖具有一定的促进作用,可能是其防治血管内皮损伤及内皮功能障碍相关性疾病的作用机制之一。Objective To investigate the effects of serum - containing Qihong decoction on proliferation of endothelial progenitor cells（EPCs）derived from peripheral blood. Methods Twelve Wistar male rats were randomly divided into four groups：Qihong decoction low dosage group, medium dosage group,high dosage group and control group. The serum -containing Qihong decoction and control serum were prepared. The mononuclear cells were isolated from human peripheral blood by density gradient centrifugation. The laser scanning confocal microscope and flow cytometry（FCM）were used to determine the EPCs in human peripheral blood. The EPCs were incubated with different serum - containing and control serum culture liquid for 24 h, 48 h and 72 h. The activity of EPCs was observed under reverse microscope. MTT was used to determine the OD value of EPCs. Results Compared with control group, the serum- contained Qihong decoction （low,medium and high dose group） promoted proliferation of EPCs （P〈0.05） with a timeeffect and dose-effect relation. The most prominent effect on proliferation for the same duration was in low dosage group （F〈0.05）. The effect to proliferation of EPCs was increased with time and reached peak at 72 h. Conclusion Serum- contained Qihong decoction could promote proliferation of the endothelial progenitor cells derived from peripheral blood. The result suggested that Qihong decoction could prevent and treat vascular endothelial injury/dysfunction - related disease.
