摘 要:目的观察健脑益智汤对拟血管性痴(VaD)大鼠的治疗作用,并探讨其机制。方法采用改进的双侧颈总动脉永久结扎法(2VO)建立VaD大鼠模型,灌胃给药,用Morris水迷宫法评价健脑益智汤对VaD模型大鼠学习记忆功能的影响,并用免疫组化法检测海马CAI区胆碱乙酰转移酶(ChAT)的表达,比较各组结果之间的差异。结果(10.6~42.4)g/kg健脑益智汤可以明显改善大鼠的学习记忆功能,健脑益智汤治疗组大鼠海马CAI区神经细胞ChAT的阳性表达明显高于模型组。结论健脑益智汤对拟VaD大鼠学习记忆功能有一定的改善作用,其机制可能与增强拟VaD大鼠海马CAI区神经细胞ChAT表达有关。Objective To study the therapeutical effect and its mechanism of Jiannaoyizhi decoction on the rats with vascular dementia (VaD). Methods The rat models with VaD were established by permanent bilateral occlusion of both common carotid arteries (2 - VO). The Jiannaoyizhi decoction were fed by intragastric administration. The protective effect of Jiannaoyizhi decoction on the learning and memory impairment was evaluated by Morris water maze techniques. The expression of choline acetyltransfrase (CHAT) in the CAI region of hippocampus was investigated by immunohistochemistry. Results Jiannaoyizhi decoction in 10.6 g/kg to 42.4 g/kg obviously improved the learning and memory ability of VaD rats. The positive expression of ChAT in the CAI region of hippocampus neurons in Jiannaoyizhi decoction group was higher than that in the model group. Conclusion Jiannaoyizhi decoction could improve the learning and memory ability in rats with VaD. The mechanism might be related to increase the expression of ChAT in the CA1 region of hippocampus neurons.
