摘  要:目的探讨体内不同环境对组织工程血管形成的影响。方法清洁级犬10只;切取一侧10 cm隐动脉,消化、离心收集内皮细胞和平滑肌细胞,与Ⅰ型胶原蛋白凝胶均匀混合,种植于猪小肠黏膜下层(small in-testinal submucosa,SIS)组织膜表面,在直径3 mm聚乙烯管壁外包绕3层,制成管状组织工程支架20支。将支架分别植于犬皮下和套于股动脉管壁外,即皮下组、动脉周围组(n=10)。术后4周,取出培养组织行大体和组织学观察,并行组织工程血管形成评分及抗静水压检测。结果皮下组管腔结构不完整,内皮及平滑肌种子细胞增殖弱,SIS支架材料分解慢,腔面无内皮细胞覆盖;动脉周围组可见完整血管样结构形成,种子细胞生长增殖良好,管腔有完整内皮细胞覆盖,平滑肌细胞形态、分布良好。皮下组、动脉周围组组织工程血管形成总评分分别为(5.529±0.272)和(14.824±0.253)分,组间差异均有统计学意义(P<0.05);抗静水压二组分别为(67.0±5.8)kPa和(247.0±3.5)kPa,动脉周围组与皮下组比较,差异均有统计学意义(P<0.01)。结论体内不同环境对细胞增殖和组织工程血管形成有明显影响。[Objective] To explore the influence of different environments on the growth of tissue engineering blood vessels in vivo. [Methods] Ten dogs of clean grade were selected, the endothelial cells and smooth muscle cells were isolated from canine saphenous artery, the seed eells were blended with type Ⅰ collagen gel, and then planted on the porcine small intestinal submueosa (SIS). Twenty three-layer tubular scaffolds were prepared with the SIS films wrapping vascular seed cells around 3 mm diameter polyethylene tubes, which were implanted into the subcutaneous tissue (subcutaneous group), and sheathed the femoral artery (perivascular group) respectively. Four weeks postoperatively, these cultured tissues were harvested, and evaluated by macroscopic observation, histology detection and anti-hydrostatic pressure detection. [Results] The cultivated tissues in different environments had obvious difference in respect of the tubular configuration, cellular proliferation and tissue shape. In subcutaneous group, the wall structure integrity, seed ceil proliferation and SIS scaffold decomposition were poor, lumen surface was eovered without endothelial eells, in perivascular group, vascular-like structure formed, the seed cell growth and proliferation were good, the lumen surface was completely covered with endothelial cells, the smooth muscle cells were in good morphological distribution, the anti-hydrostatic pressure was (247.0±3.5) kPa, showing significant differences when compared with subcutaneous group (67.0±5.8) kPa (P 〈0.01). The total scoring of tissue engineering blood vessel formation in subcutaneous group and perivascular group were (5.529±0.272), (14.824±0.253)respectively, and the differences among them were significant (P 〈0.05). [Conduslon] In vivo different environments have a great influence on the cellular proliferation and the growth of tissue engineering blood vessel.
