摘  要:日本血吸虫成虫26kD谷胱甘肽S-转移酶(Sj26KDGST)是成虫代谢过程中一个关键酶。为研究制备日本血吸虫重组疫苗,根据日本血吸虫菲律宾株成虫Sj26kDGST完整编码区序列设计引物,以日本血吸虫中国大陆株成虫26kDGSTmRNA为模板,采用逆转录-聚合酶链反应技术,扩增了其26kDGST完整编码区cDNA。将cDNA重组于pBluescriptSK质粒上,转化大肠杆菌,重组体经限制酶解图谱鉴定,用双脱氧链末端终止法对克隆的编码区CDNA的5’端和3’端大部分顺序进行测定,结果与菲律宾株成虫蜀26kDGST编码区CDNA进行了比较,表明二者顺序一致。为表达和制备血吸虫重组疫苗奠定了初步的基础。The 26kD Schistosoma japonicum glutathione S-transferase(Sj26 GST) was key enzyme in metabolism proceeding of theadult.The gene encoding Chinese mainlad strain Sj26 GST was amplified with RT-PCR by using the primer synthesized with reference to the Sj26 GST gene sequence of Philippines for research of anti-Schistosoma vaccine.The gene was inserted into plasmid pBluescript-SK and the recombinant were transformed to E.coli DH5a.A recombinant colony has been obtained ad identified by restriction enzyme。Mapping and the' and 3’ terminal cDNA sequencing,showing that gene of Sj26 GST has been inserted into the plasmid.Compared nucleotides sequence with Philippine, the Sj26 GST between the Chinese and Philippine at the nucleotides sequence level are identical.The availability of the study should facilitate the development of the recombinant vaccine of Chinese mainland strain Sj26 GST.
